Genomska analiza polimorfizmov mutant lokalnega izolata kvasovke Lachanchea thermotolerans s spremenjenim fenotipom privzema sladkorjev

Magistrska naloga raziskuje spremembe fenotipa metabolizma sladkorjev pri mutantah kvasovke Lachancea thermotolerans ki so bile analizirane na genskem nivoju L thermotolerans je fermentativna kvasovka s potencialom za izboljsanje pivskih arom in ucinkovitosti fermentacije Mutante so bile ustvarjene s tehnikami mutageneze in usmerjene evolucije kot posledica odpornosti na analog glukoze Z obdelavo nukleotidnih zaporedij mutant in divjega tipa s programom Geneious smo primerjali genome naravnega seva in na 2DOG odpornih mutant in identificirali polimorfizme posameznega nukleotida SNP Uporabljena orodja kot sta BLAST in KEGG so omogocila identifikacijo homolognih genov Saccharomyces cerevisiae Identificirali smo stevilne SNPje na razlicnih genih Nekateri so se nahajali na kodirajocih drugi pa na nekodirajocih delih genov Med drugimi smo zaznali mutacije na genih povezanih z metabolizmom sladkorjev ki bi lahko bile povezane z opazeno spremembo fenotipa posameznih mutant Pri mutantah L34 L55 in L82 je bila med drugimi zaznana mutacija na genu YGL253W HXK2 verjetni heksokinazi Mutanta L75 pa ima SNP mutacijo na genu YOL156W HXT11 verjeten transporter heksoze Mutacije SNP na genih povezanih z metabolizmom sladkorjev bi lahko prispevale k redukciji glukozne represije kar omogoca bolj ucinkovit metabolizem sladkorjev Nas pristop je omogocil identifikacijo sprememb v izbranih mutantah in s tem prispeval k razumevanju katabolne represije ter ponuja moznosti za razvoj novih pivskih kvasovk z izboljsanimi lastnostmi

We explored phenotypic changes in sugar metabolism of Lachancea thermotolerans mutants which were analyzed at the genetic level L thermotolerans is a fermentative yeast with potential for improving beer aromas and fermentation efficiency The mutants were created using mutagenesis and directed evolution approaches A number of single nucleotide polymorphisms SNPs were identified by applying comparative analysis of mutants and the wild type genomes BLAST and KEGG tools enabled the identification of homologous genes in Saccharomyces cerevisiae Some SNPs were located in coding regions while others were in non-coding regions of the genes Among others we detected mutations in genes associated with sugar metabolism which could be linked to the observed phenotypic changes in certain mutants such as resistance to 2- deoxyglucose and altered sugar metabolism In mutants L34 L55 and L82 a mutation was detected in the YGL253W HXK2 gene which encodes putative hexokinase Mutant L75 has an SNP mutation in the YOL156W HXT11 gene which encodes a putative hexose transporter SNP mutations in genes related to sugar metabolism could contribute to the reduction of glucose repression enabling more efficient sugar metabolism Our approach allowed the identification of changes in selected mutants contributing to the understanding of the genetic basis of catabolite repression during fermentation We developed new brewing yeasts strains with improved properties in sugar assimilation
