Vpliv vnetja na razporeditev kanabinoidnega receptorja 2 v urotelijskih celicah in vitro

Intersticijski cistitis (IC) je kronično vnetje sečnega mehurja, ki ga spremljajo bolečine v sečnem mehurju, spremembe v frekvenci uriniranja in nelagodje v predelu medenice. Vzroki za nastanek te bolezni do sedaj še niso znani, vendar se zaradi pogostega ponavljanja simptomov te bolezni, stremi k odkritju možnih tarč za zdravljenje. Ena izmed možnih tarč je kanabinoidni receptor 2 (CBR2), ki je prisoten v urotelijskih (epitelijskih) celicah sečnega mehurja sesalcev. V magistrski nalogi smo ugotavljali razporeditev in količino CBR2 v kulturah normalnih prašičjih urotelijskih celic (NPU) in v celičnih linijah neinvazivnega človeškega urotelijskega papiloma (RT4). Uporabili smo netretirane (kontrolne) celice in celice, tretirane s tumor nekrotizirajočim faktorjem α (TNFα), ki v celicah izzove vnetje, s čimer smo dobili in vitro model IC. Za ugotavljanje razporeditve in količine CBR2 v celicah smo uporabili imunofluorescenčno označevanja CBR2 ter za ugotavljanje količine CBR2 prenos western. Ugotovili smo, da je CBR2 v netretiranih celicah NPU prisoten v citoplazmi, v celicah NPU tretiranih s TNFα pa je CBR2 v citoplazmi in v plazmalemi. V netretiranih kot tudi v TNFα tretiranih celicah RT4 je bil CBR2 vedno prisoten le v citoplazmi. Količina receptorja se je v celicah NPU po izzvanem vnetju povečala, v celicah RT4 pa zmanjšala glede na celice brez izzvanega vnetja, vendar v obeh primerih brez statistično značilne razlike. Vzorec razporeditve CBR2 je bil v netretiranih kot tudi tretiranih celicah NPU in RT4 točkast. Rezultati magistrske naloge kažejo na razlike v odzivanju zdravih urotelijskih celic in celic urotelijskega papiloma na TNFα, kar se je izrazilo tudi v razlikah v količini in razporeditvi izraženega receptorja. Ti rezultati so pomembna osnova za nadaljnje raziskave razumevanja vloge CBR2 kot terapevtske tarče za zdravljenje IC.

Naslov:Effect of inflammation on the distribution of cannabinoid receptor 2 in urothelial cells in vitro

Interstitial cystitis (IC) is a chronic inflammation of the bladder, accompanied by pain in the bladder, changes in the frequency of urination and discomfort in the pelvic area. The causes of this disease are still unknown, but due to the frequent recurrence of the symptoms of this disease, efforts are being made to discover possible targets for treatment. One possible target is the cannabinoid receptor 2 (CBR2), which is present in the urothelial (epithelial) cells of the mammalian bladder. In my master's thesis, we determined the distribution and the amount of CBR2 in cultures of normal porcine urothelial cells (NPU) and in non-invasive human urothelial papilloma (RT4) cell lines. We used untreated (control) cells and cells treated with tumor necrosis factor α (TNFα), which induces inflammation in cells, to obtain an in vitro model of IC. CBR2 immunofluorescence labeling was used to determine the distribution and amount of CBR2 in cells, and western blotting was used to determine the amount of CBR2 in cells. We found that in untreated NPU cells, CBR2 is present in the cytoplasm, and in NPU cells treated with TNFα, CBR2 is present in the cytoplasm and in the plasmalemma. In untreated as well as TNFα-treated RT4 cells, CBR2 was always present only in the cytoplasm. The amount of receptors increased in NPU cells after induced inflammation and decreased in RT4 cells compared to cells without induced inflammation, but in both cases without statistically significant differences. The distribution pattern of CBR2 was punctate in untreated as well as treated NPU and RT4 cells. The results of my master's thesis indicate differences in the response of healthy urothelial cells and urothelial papilloma cells to TNFα, which was also expressed in differences in the amount and distribution of the expressed receptor. These results thus provide an important basis for further research to understand the role of CBR2 as a therapeutic target for the treatment of IC.
