Abstract
V diplomskem delu smo preučevali vpliv različnih hormonskih gojišč na razvoj koreninskega sistema modelne rastline Nicotiana benthamiana ter antagonistične interakcije med izbranimi glivnimi sevi. Rastline smo gojili na gojiščih z različnimi koncentracijami rastnih regulatorjev IBA, BAP in NAA ter na kontrolnem gojišču brez dodanih rastnih regulatorjev. Spremljali smo razvoj korenin, tvorbo kalusa in poganjkov ter rast rastlin. Najbolj razvit koreninski sistem so razvile rastline na kontrolnem gojišču, medtem ko je pri večini hormonskih gojišč prevladovala tvorba kalusa ali poganjkov. V drugem delu raziskave smo na PDA-gojišču spremljali medsebojno rast šestih glivnih sevov iz rodov Fusarium in Trichoderma. Antagonistične interakcije smo vrednotili na podlagi nastanka inhibicijskih oziroma demarkacijskih con in odsotnosti medsebojnega preraščanja kolonij. Opazili smo jih le pri nekaterih kombinacijah, predvsem med sevi rodu Trichoderma. Rezultati kažejo, da uporabljeni rastni regulatorji niso izboljšali razvoja korenin v primerjavi s kontrolnim gojiščem ter da so bile antagonistične interakcije odvisne od posamezne kombinacije glivnih sevov.

Title:Optimization of In Vitro Root Growth of Nicotiana benthamiana and Antagonism of Trichoderma harzianum against Rhizoctonia solani
Abstract:
This thesis examined the effects of different hormone supplemented media on root system development in the model plant Nicotiana benthamiana, as well as antagonistic interactions among selected fungal strains. Plants were grown on media containing different concentrations of the plant growth regulators IBA, BAP and NAA, and on a control medium without added growth regulators. Root development, callus and shoot formation, and overall plant growth were monitored. The most developed root system was observed in plants grown on the control medium, while callus or shoot formation predominated on most hormone-supplemented media. In the second part of the study, interactions among six fungal strains from the genera Fusarium and Trichoderma were monitored on PDA medium. Antagonistic interactions were evaluated based on the formation of inhibition zones and the absence of colony overgrowth. Such interactions were observed only in some combinations, mainly between strains of the genus Trichoderma. The results show that the tested plant growth regulators did not improve root development compared with the control medium and that antagonistic interactions depended on the specific combination of fungal strains.
