Abstract
Amiloidi so vrsta proteinov, ki tvorijo agregate v obliki fibril. Njihova sekundarna struktura je bogata z β-ploskvami. Te predstavljajo tarčno vezavno mesto za fluorescenčne molekulske sonde, ki so lahko preko biotin–streptavidin interakcije vezane na magnetne nanodelce in se med drugim uporabljajo v raziskavah zgodnje detekcije nekaterih nevrodegenerativnih bolezni. V diplomski nalogi je predstavljena vezava izbrane fluorescenčne molekulske sonde na biotin preko dietilenglikolnega vmesnika. Sinteza je potekala v več stopnjah. Iz terc-butil (2-(2-(2-hidroksietoksi)etoksi)etil) karbamata in anhidrida jantarne kisline smo najprej sintetizirali vmesnik. Nanj smo s Steglichovim estrenjem vezali izbrano fluorescenčno molekulsko sondo, (E)-2-(metil(4-(2-nitrovinil)fenil)amino)etan-1-ol, ki smo jo s Henryjevo reakcijo pripravili iz 4-((2-hidroksietil)(metil)amino)benzaldehida. Dobili smo N-Boc zaščiteno spojino, na katero smo v zadnji stopnji po odstranitvi Boc zaščite vezali še biotin, ki smo ga aktivirali s pomočjo reagenta PyBOP.

Title:Binding of diethylene glycol to a fluorescent molecular probe
Abstract:
Amyloids are a type of protein that forms fibril-like aggregates. Their secondary structure is rich in β-sheet motifs. These represent a target binding site for fluorescent molecular probes, which can be attached to magnetic nanoparticles via biotin–streptavidin interaction and used in research on early detection of certain neurodegenerative diseases. This bachelor’s thesis presents the binding of a fluorescent molecular probe to biotin via a diethylene glycol linker, which was achieved through a multiple-step approach. First, a linker was synthesized using tert-butyl (2-(2-(2-hydroxyethoxy)ethoxy)ethyl)carbamate and succinic anhydride. A fluorescent molecular probe, (E)-2-(methyl(4-(2-nitrovinyl)phenyl)amino)ethan-1-ol, prepared from 4-((2-hydroxyethyl)(methyl)amino)benzaldehyde by the Henry reaction, was then coupled to it via Steglich esterification. This yielded an N-Boc protected compound to which biotin, activated by PyBOP reagent, was bound after the removal of a Boc protecting group.
