Abstract
Živilski in lesni ostanki so odličen vir surovin za izvajanje okolju prijaznih biotehnoloških procesov. V ta namen smo v magistrski nalogi poskusili izrabiti potencial bioostankov ter z različnimi naravnimi razkrojevalci in plesnimi dobiti povečane količine sproščenih polifenolov. Polifenoli so naravne spojine, ki izkazujejo velik aplikativni potencial v živilski, farmacevtski in kozmetični industriji predvsem zaradi antioksidativne sposobnosti. Kot delovne organizme smo določili štiri vrste gliv: A. niger, R. arrhizus, P. ostreatus in A. flavus ter šest različnih substratov za izvajanje kultivacije na trdnem gojišču (SSF): uporabljene čajne vrečke metinega, zelenega in sadnega čaja, kavna usedlina ter otrobe jedrovine in skorje jelke. Kultivacijo smo uspešno zastavili in je trajala 15 dni, pri čemer smo vzorčenje izvedli dvakrat. Da bi na koncu procesa SSF dobili fenolne spojine, smo izvedli ekstrakcijo s tremi tehnikami, z magnetnim mešalom, z ultrazvokom in s pospešeno ekstrakcijo s topilom. Vzorce, ki so jih v našem primeru predstavljali polifenolni ekstrakti, smo gravimetrično in spektrofotometrično analizirali ter pregledali profil prisotnih spojin s tekočinske kromatografije visoke ločljivosti. Obdelani rezultati so pokazali, da smo uspešno izvedli proces in da lahko s primernimi delovni organizmi iz bioostankov pridobimo večje količine polifenolov. Da bi bil celotni postopek pridobivanja polifenolnih spojin učinkovitejši in ekonomsko ugodnejši, smo pregledali še, katera metoda ekstrakcije je najbolj učinkovita. Izkazalo se je, da so vse tri tehnike med seboj primerljive in učinkovite.

Title:Production of phenolic compounds by solid-state fermentation of selected biological residues using different fungal species
Abstract:
Food and wood residues are a great source of raw materials for environmentally friendly biotechnological processes. In that purpose in the master thesis, we tried to exploit the potential of bio-residues and to obtain increased amounts of released polyphenols with four fungal species, natural decomposers and molds in a solid-state cultivation (SSF) process. Polyphenols are natural compounds that show great applicative potential in food, pharmaceutical and cosmetic industry, mainly due to their antioxidant capacity. Our selected fungal species were A. niger, R. arrhizus, P. ostreatus and A. flavus and 6 different substrates were used to carry out SSF: used mint, green and fruit tea bags, coffee grounds, fir bran from cores and bark. Cultivation lasted for 15 days, with two sampling times. Extraction of the samples was achieved by 3 techniques: magnetic stirrer, ultrasound sonication and accelerated solvent extraction, simultaneously examining which is the most efficient and economically advantageous. Resulting that all are comparable and effective. The extracts were analysed gravimetrically and spectrophotometrically, the profile of the present compounds was examined by high-performance liquid chromatography. Results suggest that SSF process was successfully carried out and higher amounts of polyphenols can be extracted from the bio-residues with suitable fungi.
