Abstract
Zaradi številnih pozitivnih učinkov rastlin in njihovih izvlečkov se le-te vse pogosteje uporabljajo v vsakdanjem življenju. Navadni rožmarin (Rosmarinus officinalis) se že od nekdaj uporablja kot zdravilna rastlina in začimba, v zadnjih letih pa je svoj razcvet doživel tudi na področju medicine, farmacije in kozmetične industrije. V kozmetičnih izdelkih je rožmarin največkrat prisoten v obliki rastlinskega izvlečka, kjer ima antioksidativno, protimikrobno in okluzivno vlogo, pretežno pa deluje kot korigens vonja ali okusa. V diplomski nalogi smo ocenili varnost izvlečka navadnega rožmarina kot sestavine kozmetičnega izdelka z uporabo pristopa toksikološkega tveganja (TTC), ki na osnovi kemijskih struktur posamezne spojine uvrsti v Cramer-jeve razrede toksičnosti, katerim so pripisane specifične vrednosti TTC. S pregledom literature smo proučili kvantitativno in kvalitativno sestavo rastline in izvlečka lista, cveta in olja listov navadnega rožmarina. S programom ChemOffice smo nato izračunali nekatere fizikalno-kemijske lastnosti posamičnih sestavin, s pomočjo katerih smo tem sestavinam ocenili potencialni obseg absorpcije in posledično tudi sistemsko izpostavljenost ob uporabi kozmetičnega izdelka, ki vsebuje izvleček navadnega rožmarina. Z računalniškim programom Toxtree smo sestavine razvrstili v Cramer-jeve razrede toksičnosti. Dobljene rezultate sistemske izpostavljenosti smo primerjali z vrednostmi, ki so pripisane posameznim Cramer-jevim razredom. Kljub visokim vrednostim sistemske izpostavljenosti v primeru obeh triterpenoidov, ki sta uvrščena v Cramer-jev razred III, izvlečka nismo določili kot toksičnega, saj so vrednosti TTC postavljene zelo nizko, kar pomeni, da majhen presežek ne predstavlja nevarnosti. Ostale izračunane vrednosti sistemske izpostavljenosti preučevanih spojin niso presegle dopustne vrednosti TTC. Nato smo s pregledom literature za posamezne spojine poiskali najvišji odmerek, pri katerem še ne pride do neželenih učinkov (NOAEL) in iz dobljenih vrednosti izračunali varno dnevno izpostavljenost. Te rezultate smo primerjali z rezultati, pridobljenimi s pristopom TTC, in ugotovili, da jih krepko presegajo.

Title:Safety assessment of rosemary (Rosmarinus officinalis) extract as a cosmetic ingredient using the threshold of toxicological concern approach
Abstract:
Due to the many positive effects of plants and their extracts, they are increasingly used in everyday life. Rosemary (Rosmarinus officinalis) has been used as a medicinal herb and spice since ancient times, and in recent years it has also made its mark in the healthcare, pharmaceutical and cosmetic industries. In cosmetic products, rosemary is most often present in the form of a plant extract, where it has antioxidant, antimicrobial and occlusive roles, and predominantly as a flavour and fragrance modifier. In this thesis, the safety of rosemary extract as a cosmetic ingredient was assessed using a threshold of toxicological concern (TTC) approach, which classifies individual compounds into Cramer toxicity classes based on their chemical structures and thus assigning them with specific TTC. The quantitative and qualitative composition of the plant and the extract of the leaf, flower and leaf oil of common rosemary were examined by literature review. We then used ChemOffice to calculate some physicochemical properties of the individual ingredients to estimate the potential extent of absorption and, consequently, the systemic exposure of these ingredients when using a cosmetic product containing rosemary extract. The Toxtree software was used to classify the ingredients into Cramer toxicity classes. The resulting systemic exposure results were compared with the values assigned to each Cramer's class. Despite the high predicted systemic exposure for both triterpenoids, classified as Cramer Class III, we have not determined the extract to be toxic, as the TTC values are set very low, meaning that a small excess does not pose a hazard. The calculated systemic exposures fort he res tof the studied compounds did not exceed the TTC values. We then searched the literature to find the no-observed-adverse-effect level (NOAEL) for each compound from which safe daily exposures were calculated. The latter were compared with the results obtained using the TTC approach, which led to the conclusion that safe daily exposures by far superseed those generated by the TTC approach.
