Izražanje izbranih molekul mikro RNA v koži bolnikov z vaskulitisom povzročenim z IgA

Abstract
Vaskulitis povzročen z IgA (IgAV) je avtoimunska bolezen, v žilah se odlagajo imunoglobulini IgA. Gre za sistemsko vnetje, ki prizadene male krvne žile v koži, ledvicah in črevesju. Vzrok bolezni še ni odkrit. Nekodirajoče RNA, med katere sodijo mikro RNA (miRNA) so izredno pomemben dejavnik v uravnavanju izražanja genov na post-transkripcijskem nivoju. Spremembe v njihovem izražanju prispevajo k nastanku bolezni. Namen dela je bila analiza izražanja mikro RNA miR-17-5p, miR-21-3p, miR-21-5p in miR-451a v 64 vzorcih kože odraslih bolnikov z IgAV in 19 odraslih zdravih posameznikov. Raven izražanja izbranih miRNA smo merili s kvantitativnim PCR v realnem času in z relativno kvantifikacijo primerjali izmerjene ravni njihovega izražanja v posameznih vzorcih. Z Mann-Whitney U testom smo razlike opredelili kot statistično pomembne in preverili morebitne povezave s kliničnimi znaki in podatki patoloških analiz. Dokazali smo statistično značilno spremenjeno izražanje vseh štirih izbranih mikro RNA v koži bolnikov v primerjavi s kožo zdravih, od katerih je bila miR-21-3p najbolj izrazito prekomerno izražena. Zaradi statistično značilnega različnega izražanja ob primerjavi histopatološke infiltracije z granulociti, sklepamo o povezavi miR-17-5p z zagonom bolezni, na podlagi statistično pomembnih razlik pri bolnikih s klinično opredeljeno nekrozo kože ter gastrointestinalno okvaro, pa sklepamo na povezavo mir-21-3p s hujšo obliko bolezni, ki jo lahko zaznamujeta kroničen potek in izrazitejša prizadetost organov.

Title:Differential expression of selected microRNA molecules in affected skin of patients with IgA vasculitis
Abstract:
IgA vasculitis (IgAV) is an autoimmune disease, characterized by IgA antibody deposits in vessel walls. It is defined as a systemic inflammation of small vessels found in skin, kidney and gastrointestinal tract . The cause of IgAV is yet to be revealed. Noncoding RNAs, micro RNAs specifically, play a crucial role in post-transcriptional regulation of gene expression. Therefore, a shift in their own expression promotes disease onset. The objective of our study was to analyse differential expression of micro RNAs miR-17-5p, miR-21-3p, miR-21-5p and miR-451a in affected skin of 64 IgAV patients and 19 healthy individuals. Using the quantitative real-time PCR method and relative quantification along with Mann-Whitney U test, we determined statistically significant differential expression of all four micro RNAs, with miR-21-3p being the most prominently up-regulated in IgAV patients. When correlating our figures with IgAV patients clinical data and parameters of pathological analysis, we found statistical significance of miR-17-5p expression in level of granulocyte infiltration and statistical significance of miR-21-3p expression in relation to skin necrosis and gastrointestinal tract involvement. Accordingly, we infer role of miR-17-5p in the earliest stages of IgAV, while miR-21-3p could be associated with chronic condition.
