Abstract
Pri diplomskem delu nas je zanimalo, ali se fluorescenčno označena egerolizinska proteina, ostreolizin A in erilizin A, vežeta na kulturo diferenciranih živčnih celic ter na celice človeške ustne sluznice. Ugotovili smo, da se na kulturo človeških živčnih celic veže le fluorescenčno označen ostreolizin A, kar sovpada z dejstvom, da se ostreolizin A specifično veže na lipidne domene, obogatene s holesterolom in sfingomielinom, ki so v teh celicah prisotne. Vezava fluorescenčno označenega erilizina A na isti celični tip je bila zanemarljivo šibka oziroma je ni bilo. Prav tako smo na in vitro modelu celic človeške ustne sluznice zasledili le vezavo fluorescenčno označenega ostreolizina A, ne pa tudi erilizina A, ki se na celice človeške ustne sluznice ni vezal.

Title:Binding of fluorescently tagged egerolysin proteins ostreolysin A and erylysin A on in vitro culture of differentiated brain cells and on human mucous membrane cells
Abstract:
In this work, we were interested in whether the fluorescently-labeled egerolysin proteins, ostreolysin A and erylsin A, bind to culture of differentiated nerve cells and to the human oral mucosal cells. We showed that fluorescently-labelled ostreolysin A binds to human nerve cells, since these cells contin lipid domains rich in cholesterol and sphingomyelin. On the other hand, erylysin A did not bind to this cell line. Simillary, fluorescently-tagged ostreolysin A binds to human oral mucosal cells, but the binding of fluorescently-tagged erilysin A was not observed.
