Abstract
DNaza je encim, ki ga uvrščamo med endonukleaze. To je najbolj preučeni encim med endonukleazami, ki razgrajuje dvoverižno DNA. Poleg navedene funkcije se DNaza uporablja tudi kot orodje v molekularni biologiji in, tudi pri zdravljenju cistične fibroze. Citotoksini CdtABC so skupina proteinskih toksinov, ki so prisotni pri bakterijah iz družine Proteobacteria. Zaradi DNazne aktivnosti so genotoksični za evkariontske celice V svoji diplomski nalogi sem primerjala aktivnost encima iz skupine endonukleaz, to jegovejo pankreasno endonukleazo DNazo I s podenoto CdtB iz bakterije Aggregatibacter actinomycetemcommitans. Osredotočila sem se na primerjavo dveh encimskih metod,in na pogoje, ki so potrebni za aktivnost DNaze I in podenote B iz citotoksina CdtABC.

Title:DNase activity of DNase I from bovine pancreas and B subunits of the bacterial cytotoxin CdtABC
Abstract:
DNase is an enzyme belonging to the endonuclease group. It is the most examined enzyme among endonucleases that degrades double-stranded DNA. In addition, DNase is used as a tool in molecular biology and for treating cystic fibrosis. Cytotoxins CdtABC are a group of protein toxins that are present in bacteria from the Proteobacteria family. Owing to their DNase activity, they are genotoxic to eukaryotic cells. The thesis compares the activity of an enzyme from the endonuclease group, i.e. bovine pancreatic endonuclease DNase I with the CdtB subunit from the bacteria Aggregatibacter actinomycetemcommitans. We focused on the comparison of two enzymatic methods and the conditions necessary for the activity of DNase I and subunit B from the CdtABC cytotoxin.
