Abstract
Osnovna funkcija celične membrane je ločevanje celične notranjosti od okolice. Zaradi ionskega neravnovesja med celično notranjostjo in okolico je na celični membrani prisotna membranska napetost. Prehod ionov preko celične membrane uravnavajo ionski kanalčki, ki so večinoma napetostno odvisni in na katere lahko vplivamo s spremembo membranske napetosti. Celična membrana je kompleksna struktura, zato želimo z enostavnejšimi strukturami kot je ravninski lipidni dvosloj opisati dogajanje, ki se dogaja v celični membrani. Njegova prednost je, da je dostopen iz obeh strani, lahko pa ga tudi modeliramo kot električno vezje paralelno vezanega upora in kondezatorja. V svoji diplomski nalogi sem preučil sisteme, ki se že uporabljajo za opazovanje ionskih kanalčkov ter načrtal izgradnjo novega sistema. Za merjenje aktivnosti ionskih kanalčkov na ravninskem lipidnem dvosloju potrebujemo napetostni generator in merilnik.

Title:
Measurement device for detecting ion channels activity enbeded in planar lipid bilayers
Abstract:
The cell is the basic structural biological unit known to all living organisms. All living cells have between interior and exterior difference of potentials, called membrane potential. This membrane potential depends mainly on the activity of ion channels. Ion channels are transmembrane proteins that are selectively permeable to the ions. They have important role in regulation of cellular ionic homeostasis, and they participate in communication between the cells and environment. Change of membrane voltage have impact on activity of voltage gated ion channels.. Artificial membranes where there are no active processes and where we have access to the both side of membranes are best way to study ion channels. This thesis represents device which can impose voltage to the cell membrane and cause the activity of ion channels. Device measure the current of ions through ion channels and convert it into voltage signal that can be observed on oscilloscope.
