Antagonistični potencial gliv Trichoderma spp. proti izbranim talnim fitopatogenim glivam

Abstract
Namen magistrskega dela je bil ovrednotiti antagonistični potencial izbranih izolatov gliv Trichoderma spp. proti izbranim fitopatogenim talnim glivam Agroathelia rolfsii, Sclerotinia sclerotiorum in Sclerotium cepivorum pri temperaturah 15 in 25 °C. V poskusu smo uporabili izolate T. atroviride in T. afroharzianum iz pripravkov Tricho Immun in Vintec ter lokalne izolate KIS 1872 in KIS 2885 istih vrst iz mikološke zbirke Kmetijskega inštituta Slovenije ter jih preizkusili v testu dvojnih kultur in vitro. Rast micelija smo spremljali deset dni, izmerili površino kolonij, izračunali delež inhibicije rasti ter vizualno ocenili interakcije med antagonisti in patogeni. Rezultati so pokazali, da so vsi izolati Trichoderma spp. pri 25 °C rasli hitreje kot pri 15 °C; pri 25 °C so vsi prerasli agarne plošče četrti dan po inokulaciji, pri 15 °C pa je bila posebej slaba rast izolata T. afroharzianum iz pripravka Tricho Immun. Izolati so večinoma statistično značilno zmanjšali rast patogenov glede na kontrole. Pri 25 °C so bili deleži inhibicije med izolati primerljivi, pri 15 °C pa je bil najučinkovitejši izolat T. atroviride iz pripravka Vintec, najmanj učinkovit pa izolat T. afroharzianum iz pripravka Tricho Immun. Največja inhibicija je bila ugotovljena pri patogenu S. sclerotiorum, zlasti pri 25 °C. Kot pokazatelj vpliva antagonistov na razvoj patogenov smo spremljali tvorbo sklerocijev in ugotovili, da je gliva A. rolfsii sklerocije tvorila le pri 25 °C, največ na kontrolnih ploščah, S. sclerotiorum pa le na kontroli pri 25 °C. Pri S. cepivorum so bili sklerociji prisotni v vseh obravnavanjih. Rezultati potrjujejo, da je antagonistični učinek izolatov Trichoderma spp. odvisen od temperature, patogene vrste in posameznega izolata.

Title:Antagonistic potential of Trichoderma spp. against selected soil-borne phytopathogenic fungi
Abstract:
The aim of this master’s thesis was to evaluate the antagonistic potential of selected Trichoderma spp. isolates against selected soil-borne phytopathogenic fungi Agroathelia rolfsii, Sclerotinia sclerotiorum and Sclerotium cepivorum at 15 and 25 °C. The isolates T. atroviride and T. afroharzianum from Tricho Immun, Vintec, and local isolates KIS 1872 and KIS 2885 from mycological collections of Agricultural Institute of Slovenia, were tested using a dual culture assay. Mycelial growth was monitored for ten days, colony area was measured, growth inhibition was calculated, and interactions between antagonists and pathogens were visually assessed. The results showed that all Trichoderma spp. isolates grew faster at 25 °C than at 15 °C; at 25 °C all isolates overgrew the agar plates by the fourth day after inoculation, whereas at 15 °C the isolate T. afroharzianum from the Tricho Immun product showed particularly poor growth. In most treatments, the isolates significantly reduced pathogen growth compared to the controls. At 25 °C, inhibition levels were relatively similar among isolates, while at 15 °C the isolate T. atroviride from Vintec was the most effective and isolate T. afroharzianum from Tricho Immun the least effective antagonist. The highest inhibition was observed against S. sclerotiorum, especially at 25 °C. Sclerotia formation was also monitored as an indicator of the effect of antagonists on pathogen development. A. rolfsii formed sclerotia only at 25 °C, with the highest number on control plates, while S. sclerotiorum formed sclerotia only on the control plate at 25 °C. In S. cepivorum, sclerotia were present in all treatments. The results confirm that the antagonistic activity of Trichoderma spp. isolates depends on temperature, pathogen species and the individual isolate.
