Optimizacija postopkov za pridobivanje terapevtskih proteinov iz transgenih rastlin

Abstract
Namen diplomskega dela je bil preučiti uporabo transgenih rastlin za proizvodnjo terapevtskih proteinov ter predstaviti ključne postopke optimizacije, ki vplivajo na uspešnost njihove proizvodnje. Delo temelji na analizi znanstvene literature s področja molekularnega kmetijstva, rastlinskih ekspresijskih sistemov in sodobnih biotehnoloških pristopov za pridobivanje biofarmacevtikov. Obravnavane so bile metode pridobivanja transgenih rastlin, različni ekspresijski sistemi ter strategije za povečanje izražanja, stabilnosti, funkcionalnosti in izkoristka terapevtskih proteinov. Ugotovljeno je bilo, da prehodni ekspresijski sistemi omogočajo hitro in visoko učinkovito proizvodnjo proteinov, medtem ko stabilna ekspresija zagotavlja dolgoročno in stroškovno ugodno proizvodnjo. Med najpomembnejšimi dejavniki, ki vplivajo na uspešnost proizvodnje, so optimizacija ekspresijskih kaset, uporaba ustreznih promotorjev, kodonska optimizacija, glikoinženiring, usmerjanje proteinov v izbrane celične predele ter zaščita pred proteolitično razgradnjo. Analiza je pokazala tudi velik pomen nadzorovanih rastnih pogojev in sodobnih metod izolacije ter čiščenja proteinov, ki pomembno vplivajo na končni izkoristek procesa. Sklenemo lahko, da rastlinski produkcijski sistemi predstavljajo varno, prilagodljivo in stroškovno učinkovito alternativo tradicionalnim ekspresijskim sistemom, nadaljnji razvoj optimizacijskih pristopov pa bi še povečal njihov potencial za industrijsko proizvodnjo terapevtskih proteinov.

Title:Optimization of therapeutic protein production from transgenic plants
Abstract:
The aim of this bachelor's thesis was to review the use of transgenic plants for the production of therapeutic proteins and to present the main optimization strategies that affect the efficiency of their production. The thesis is based on a full analysis of scientific literature related to molecular farming, plant expression systems and modern biotechnological approaches for the production of biopharmaceuticals. Different methods for generating transgenic plants, various expression systems and strategies for improving the expression, stability, functionality and yield of therapeutic proteins were examined. The analysis showed that transient expression systems provide a cost-effective platform for long-term production. Key factors affecting production efficiency include optimizing expression cassettes, using suitable promoters, codon optimization, glycoengineering, subcellular targeting of proteins and protection against proteolytic degradation. The review also highlighted the importance of controlled cultivation conditions and advanced downstream processing methods, which significantly affect final product yield and quality. It can be concluded that plant-derived production platforms present a safe, flexible and economically attractive alternative to conventional expression systems and that further advances in optimization strategies will improve their potential for large-scale industrial production of therapeutic proteins.
