Toplotna odpornost proteaz bakterij Pseudomonas spp. v homogenatu in svežem piščančjem mesu

Abstract
V tem delu smo preučevali proteazno aktivnost izolatov bakterij Pseudomonas spp. in toplotno odpornost njihovih proteaz v različnih medijih. Proteazno aktivnost smo analizirali z azokazeinsko metodo po inkubaciji izolatov v triptičnem sojinem bujonu, homogenatu piščančjega mesa in svežem piščančjem mesu. Ohranjeno proteazno aktivnost smo analizirali po 5- in 30-minutni toplotni obdelavi gojišča pri 50, 60, 70, 80 in 90 °C. Izolati so se razlikovali v proteazni aktivnosti in toplotni odpornosti proteaz, ki sta bili odvisni od rastnega gojišča ter časa in temperature inkubacije med rastjo in toplotno obdelavo. Visoka celična gostota ni vedno pomenila prisotnosti proteazne aktivnosti. Proteazno aktiven je bil tudi izolat CC135, pri katerem prisotnost operona aprX-lipA2 ni bila potrjena. V tekočih gojiščih TSB in homogenatu piščančjega mesa so proteaze po toplotni obdelavi ohranile del aktivnosti, pri čemer ohranjena aktivnost ni padala linearno z višanjem temperature, ampak je izkazovala pojav inaktivacije proteaz pri nižjih temperaturah toplotne obdelave. V svežem piščančjem mesu smo po 7 dneh inkubacije zaznali proteazno aktivnost, vendar je po 30-minutni toplotni obdelavi nismo več zaznali v nobenem vzorcu. S cimogramom smo aktivne proteaze ocenili na molekulsko maso 50 kDa. Pokazali smo, da na podlagi rezultatov o proteazni aktivnosti in toplotni odpornosti proteaz bakterij Pseudomonas, pridobljenih v tekočih laboratorijskih gojiščih, ne moremo neposredno sklepati o njihovem obnašanju v trdnih substratih, kot je sveže piščančje meso, saj se njihova aktivnost in odziv na toplotno obdelavo razlikujeta glede na matriks.

Title:Heat resistance of Pseudomonas spp. proteases in homogenate and fresh chicken meat
Abstract:
In this work, we studied the protease activity of Pseudomonas spp. bacterial isolates and the heat resistance of their proteases in different media. Proteolytic activity was determined by the azocasein method after incubation of isolates in tryptic soy broth, chicken meat homogenate and fresh chicken meat. Residual proteolytic activity was determined after 5 and 30 minutes of heat treatment at different temperatures. Isolates differed in proteolytic activity and heat resistance of their proteases, which depended on the growth medium and the time and temperature of incubation during growth and heat treatment. High cell density did not always indicate the presence of protease activity. Isolate CC135 was also proteolytically active, even though the presence of the aprX-lipA2 operon was not confirmed for it. In liquid media, proteases retained part of their activity after heat treatment, with the residual activity not only decreasing linearly with increasing temperature, but rather showing the phenomenon of inactivation at lower temperatures. Protease activity was detected in fresh chicken meat after 7 days of incubation, but was no longer detected after heat treatment. The active proteases were estimated by zymogram to be 50 kDa. We have shown that based on the results of protease activity and heat resistance of Pseudomonas proteases, obtained in liquid laboratory media, we cannot directly conclude about their behavior in solid substrates, such as fresh chicken meat, since their activity and response to heat treatment vary depending on the matrix.
