Abstract
Spremljanje kakovosti naravnih kopalnih voda temelji predvsem na določanju fekalnih indikatorskih bakterij (FIB), zlasti bakterije Escherichia coli, vendar sama številčnost bakterij zagotavlja le omejene informacije o virih onesnaženja, virulentnem potencialu in odpornosti proti protimikrobnim učinkovinam (AMR). V raziskavi smo z uporabo integriranega pristopa, ki je vključeval kultivacijske, fenotipske in filogenetske metode ter analizo celokupne skupnostne DNA, preučevali prostorsko in sezonsko dinamiko bakterije E. coli v Blejskem jezeru in njegovih pritokih v obdobju enega leta. Številčnost bakterije E. coli je bila v pritokih stalno višja kot v jezerski vodi in se je povečala v obdobjih, povezanih s kmetijskim gnojenjem in kopalno sezono. Filogenetska analiza je pokazala prevlado okoljsko prilagojenih filogenetskih skupin A in B1. Z virulentnostjo povezani geni so bili široko razširjeni, pri čemer je bil gen fimH zaznan skoraj pri vseh izolatih. Fenotipska odpornost proti protimikrobnim učinkovinam je bila na splošno nizka, najvišji delež odpornosti pa je bil ugotovljen proti ampicilinu (19,5 %) in tetraciklinu (13,8 %) ter je bil značilno pogostejši med izolati iz pritokov in izven kopalne sezone. Nasprotno je analiza celokupne skupnostne DNA pokazala raznolik rezistom na celotnem porečju, pri čemer sta bili raznolikost in številčnost genov za odpornost proti protimikrobnim učinkovinam (ARG) večji na mestih dotoka, gen intI1 pa je bil široko razširjen. Zaznali smo tudi več genov za odpornost proti β-laktamom, tetraciklinom, sulfonamidom in aminoglikozidom. Posebej pomembna je bila zaznava gena za odpornost proti kolistinu mcr-4 v več kot 25 % vzorcev. Rezultati kažejo, da so pritoki pomembne vstopne točke fekalnega onesnaženja, determinant virulentnosti in odpornosti proti protimikrobnim učinkovinam v Blejsko jezero, medtem ko kmetijski in rekreacijski pritiski vplivajo na sestavo populacij bakterije E. coli. Kljub odlični kakovosti kopalne vode, ocenjeni na podlagi spremljanja FIB, naši rezultati razkrivajo širši in v veliki meri nezaznan rezervoar klinično pomembnih mikrobnih lastnosti ter podpirajo potrebo po celovitem spremljanju naravnih sladkovodnih ekosistemov, namenjenih rekreaciji.

Title:Population structure, virulence gene profiles and antimicrobial resistance of Escherichia coli from Alpine Lake Bled and its tributaries : research data underlying the article
Abstract:
Monitoring of natural recreational bathing waters relies primarily on fecal indicator bacteria (FIB), particularly Escherichia coli, but bacterial abundance alone provides limited information on contamination sources, virulence potential and antimicrobial resistance (AMR). Here, we investigated the spatial and seasonal dynamics of E. coli in Lake Bled, Slovenia, and its tributaries over a one-year period using an integrated culture-based, phenotypic, phylogenetic and total community DNA approach. E. coli abundance was consistently higher in tributaries than in lake water and increased during periods associated with agricultural fertilization and the bathing season. Phylogenetic analysis revealed a predominance of environmentally adapted phylogroups A and B1. Virulence-associated genes were widespread, with fimH detected in nearly all isolates. Phenotypic AMR was generally low, with the highest resistance observed to ampicillin (19.5%) and tetracycline (13.8%), and significantly more prevalent among tributary and off-season isolates. In contrast, total community DNA analysis demonstrated a diverse resistome across the catchment, with higher ARG diversity and abundance in inflow sites, widespread detection of intI1, and multiple β-lactam, tetracycline, sulfonamide and aminoglycoside resistance genes. Notably, the colistin resistance gene mcr-4 was detected in more than 25% of samples. The findings demonstrate that tributaries represent important entry points for fecal contamination, virulence determinants and AMR into Lake Bled, while agricultural and recreational pressures shape the composition of E. coli populations. Despite excellent bathing water quality based on FIB monitoring, our results reveal a broader and largely undetected reservoir of clinically relevant microbial traits, supporting the need for integrated surveillance of recreational freshwater ecosystems.
