Abstract
Epigenetsko pomlajevanje z delnim reprogramiranjem celic omogoča obnovo mladostnega epigenetskega profila ob ohranjanju celične identitete. Temelji na reverzibilni preureditvi epigenetskih značk, med katerimi imajo ključno vlogo metilacija DNA, posttranslacijske modifikacije histonov in reorganizacija kromatina. Postopna izguba epigenetske stabilnosti je značilen spremljevalec staranja, saj vodi v upad celične funkcionalnosti in zmanjšanje regenerativnega potenciala tkiv. Delno reprogramiranje omogoča vzpostavitev prvotne epigenetske krajine, kar prispeva k zmanjšanju fenotipskih znakov staranja ter izboljšanju celičnih funkcij, ne da bi pri tem prišlo do popolne dediferenciacije celic. Na področju pomlajevanja z delnim reprogramiranjem se odpirajo možnosti razvoja novih terapevtskih pristopov, usmerjenih v upočasnjevanje staranja in podaljševanje obdobja vitalnosti. Kljub velikemu potencialu pa ostajajo odprta številna vprašanja glede dolgoročnih učinkov delnega reprogramiranja, etičnih vidikov, varnosti in klinične uporabnosti.

Title:Epigenetic rejuvenation by partial cell reprogramming
Abstract:
Epigenetic rejuvenation through partial cell reprogramming enables the restoration of a youthful epigenetic profile while preserving cellular identity. It is based on the reversible remodelling of epigenetic marks, with DNA methylation, post-translational histone modifications, and chromatin reorganisation playing a crucial role. The gradual loss of epigenetic stability is a characteristic feature of ageing and leads to a decline in cellular functionality and a reduction in the regenerative potential of tissues. Partial reprogramming allows partial restoration of the original epigenetic landscape, reducing phenotypic signs of ageing and improving cellular functionality without complete dedifferentiation. Due to these properties, this approach opens up possibilities for the development of therapeutic strategies aimed at slowing down ageing and extending the healthspan. However, important questions remain regarding long-term effects, ethical considerations, safety, and clinical applicability.
