Abstract
This thesis investigates the possibility to make a cold plasma generator that would reduce microbial count while being able to process large amounts of wastewater. We used E. coli and B. subtilis as model microorganisms to determine the effect of cold plasma on bacteria. Using two designs, the first being a dielectric barrier discharge (DBD) with a glass vessel to hold the sample, and the second, a stream of liquid flowing through plasma in DBD, we exposed bacteria in liquid media to cold plasma. We then determined the concentration of live bacteria (CFU) by counting colonies on Lysogeny broth (LB) agar plates. Results show 40% average decrease in CFU at 90 s and 63% at 300 s and 600 s of plasma exposure in the first generator and 60% at flow 1 mL/s in the second generator. We calculated volume capacities and energy efficiencies of each generator in different configurations and determined that the second generator at flow rate of 1 mL/s is the best design.

Title:Razvoj generatorja hladne plazme za obdelavo odpadne vode
Abstract:
Ta magistrska naloga raziskuje možnost izdelave generatorja hladne plazme, ki bi zmanjšal število mikrobov, obenem pa bi bil sposoben obdelati velike količine odpadne vode. Kot modelna mikroorganizma za določitev vpliva hladne plazme na bakterije smo uporabili E. coli in B. subtilis. Za izpostavitev bakterij v tekočem mediju hladni plazmi smo zasnovali dva različna prototipa generatorjev. Prvi generator predstavlja dielektrično barierno razelekritev (DBD) s stekleno posodo za zadrževanje vzorca, drugi generator pa tok odpadne vode, ki teče skozi plazemsko razelektritev v konfiguraciji DBD. Koncentracije kolonijskih enot (CFU) smo določili s štetjem kolonij, gojenih na agar ploščah (lizogenih gojiščih). Rezultati kažejo povprečno 40 % zmanjšanje CFU po 90 s in 63 % zmanjšanje CFU po 300 s oz. 600 s izpostavljenosti plazmi v prvem generatorju. V drugem generatorju je bilo doseženo 60 % zmanjšanje CFU pri pretoku 1 mL/s. Izračunali smo volumske kapacitete in energetsko učinkovitost vsakega generatorja v različnih konfiguracijah ter ugotovili, da je uporaba drugega generatorja pri pretoku vzorca 1 mL/s najbolj smotrna.
