Abstract
Raziskave smo opravili na 6 odraslih drevesih smreke (Picea abies (L.) Karst.) na rastišču pri Panški reki v bližini Ljubljane, pri katerih smo spremljali kambijevo aktivnost, sezonsko dinamiko nastajanja ksilema in floema (rast branike in diferenciacijo celic). Iz mikro izvrtkov, odvzetih v tedenskih intervalih v rastni sezoni 2012, smo pripravili prečne prereze tkiv, ki smo jih opazovali pod svetlobnim mikroskopom in analizirali s pomočjo sistema za analizo slike. Za vsak odvzem smo prešteli število trenutno oblikovanih ksilemskih, floemskih in kambijevih celic in izmerili širine tkiv. Ugotovili smo, da se je kambijeva aktivnost pri 5 raziskanih smrekah začela 3.4.2012, pri eni pa en teden kasneje. Zaključek kambijeve celične produkcije, ki je v povprečju trajala 144 dni, smo zabeležili med 13.8. in 3.9.2012. Pri spremljanju sezonske dinamike nastajanja ksilemske branike smo ugotovili, da so prve celice v fazi površinske rasti, v povprečju nastale 17.4.; prve celice v fazi odlaganja in lignifikacije večslojne sekundarne celične stene 7.5; in prve popolnoma oblikovane celice 24.5.2012. Proučili smo tudi sezonsko dinamiko nastajanja floemske branike, kjer smo zabeležili diferenciacijo prvih celic ranega floema 20.3., pojav aksialnega parenhima 2.5. in nastanek prvih celic kasnega floema 6.5.2012. Primerjave ksilemskih in floemskih branik, nastalih v istem letu, so pokazale, da so bile floemske branike v povprečju 7,8-krat ožje od ksilemskih.

Title:Wood and phloem formation in Norway spruce at Panška reka near Ljubljana : M. Sc. thesis
Abstract:
Cambial activity and seasonal dynamics of xylem and phloem formation (tree ring increment and differentiation of cells) taken from 6 mature trees of Norway spruce (Picea abies (L.) Karst.) growing at a site near Panška reka close to Ljubljana, were monitored. Cross-sections of tissues, prepared from micro-cores, taken weekly in the growing season 2012, were observed under a light microscope and analysed using the image analysis system. The numbers of currently formed xylem, phloem and cambial cells were counted for each sampling, and the widths of the tissues were measured. Cambial activity in 5 Norway spruces started on April 3, 2012 and in 1 spruce a week later. An average completion of cambial cell production lasted 144 days, from August 13 to September 3, 2012. In seasonal formation of xylem tree rings we found out the first cells in the phase of extension growth, on average on April 17, the first cells in the phase of deposition and lignification of the multilayer secondary cell wall on May 7, while the first fully mature cells were observed on May 24. In seasonal dynamics of phloem ring formation, differentiation of the first cells of early phloem was recorded on March 20, appearance of axial parenchyma on May 2, while the formation of the first cells of late phloem on May 6, 2012. The comparison of xylem and phloem tree rings, formed in the same year, showed that phloem tree rings were on average 7-8 times narrower than the xylem ones.
