Abstract
Na krasu, ki predstavlja 40 % površin v Sloveniji in je pomemben vir pitne vode, večina površinskih vod ponikne v podzemlje. Na podlagi sekvenciranih pomnoženih delov ribosomalnih genov (16S rRNA za bakterije in arheje, 18S rRNA za evkarionte in regije ITS za glive) smo analizirali mikrobno združbo različnih kraških vod v Sloveniji in vpliv predhodne namnožitve celokupne DNA na zaznano sestavo mikrobnih združb. Večjih razlik med vodami različnih kraških jam ni bilo. V arhejski združbi so prevladovali redovi Woesearchaeales, Nitrososphaerales in Nitrosopumilales. V bakterijskih združbah so pomemben delež predstavljali rodovi Limnohabitans, Rhodoferax, Flavobacterium in Pseudomonas. Identificirani glivni rodovi so bili prisotni v pomembnih deležih le v enem ali dveh vzorcih, v ostalih niso bili prisotni oz. so bili precej bolj redki. To so bili rodovi Stereum, Betamyces, Trametes, Rhizophydium in Mortierella. Med ostalimi evkariontskimi skupinami so bili pogosti migetalkarji (Ciliophora) in zlatorjave alge (Chrysophyceae). Zaznali smo tudi členonožce (Arthropoda) in sicer red Cyclopoida. Številčnost vrst vseh združb je bila v primeru izvedbe predhodne namnožitve celokupne DNA nižja. Enako velja za pestrost, razen v primeru evkariontske združbe, kjer je bila ta v vzorcih z izvedeno predhodno namnožitvijo višja. Statistično pomembne razlike so bile prisotne le v primeru številnosti in pestrosti vrst glivne združbe ter pestrosti arhejske združbe (p ⡤ 0,05).

Title:Microbial community analysis of carst cave waters using high-throughput sequencing
Abstract:
On karst, which represents 40% of the surface in Slovenia and is an important source of drinking water, most of the surface water disappears underground. Using high‐throughput rRNA gene amplicon sequencing (bacterial and archaeal 16S rRNA, eukaryotic 18S rRNA and fungal ITS region), we analysed the microbial community of various karst waters in Slovenia and the effect of total DNA pre-amplification on the detected microbial diversity and community composition. There were no major differences between the waters of the various karst caves. The archaeal community was dominated by the orders Woesearchaeales, Nitrososphaerales and Nitrosopumilales. The genera Limnohabitans, Rhodoferax, Flavobacterium and Pseudomonas represented an important proportion of the bacterial communities. Identified fungal genera represented important proportions in only one or two samples, while in others they were absent or much rarer. These were the genera Stereum, Betamyces, Trametes, Rhizophydium and Mortierella. Among other eukaryotic groups, ciliates (Ciliophora) and golden-brown algae (Chrysophyceae) were common. We also detected arthropods (Arthropoda), namely the Cyclopoida order. When pre-amplification of total DNA was used, microbial richness was lower. The same applies to the diversity, except in the case of the eukaryotic community, where diversity was higher in the samples with pre-amplified DNA. Statistically significant differences were observed only in species richness and diversity of the fungal community and diversity of the archaeal community (p ⡤ 0.05).
