Abstract
Kalorimetrija je metoda pri kateri merimo toplotne effekte s pomočjo kalorimetra. Pri diferenčni dinamični kalorimetriji (DSC) merimo toplotni tok, ki prehaja v vzorec ali iz njega. V moji diplomski nalogi sem prikazala uporabo DSC pri analizi biopolimerov in uporabila DSC za študij stabilnosti biopolimerov. Na modelno neodvisen in modelno odvisen način sem izračunala entalpijo denaturacije za protein eritropoetin v raztopini fosfatnega pufra pH=7,2. Izračunala sem razliko v dobljenih vrednostih med obema modeloma, ter z izračunom pokazala, da protein lahko opišemo kot reverzibilen prehod med dvema stanjema. Rezultati mojega izračuna so primerljivi z rezultati iz literature.

Title:USING DIFFERENTIAL SCANNING CALORIMETRY IN THE STUDY OF BIOPOLYMER SOLUTIONS
Abstract:
Calorimetry is a method for measurement of heat effects using calorimetric instruments. Differential dynamic calorimetry (DSC) measures the heat flow going into or out of the sample cell. In my thesis, I demonstrated the use of DSC in the analysis of biopolymers and investigated the applicability of DSC in studying the stability of biopolymers. I calculated the enthalpy of denaturation for the protein erythropoietin in a phosphate buffer solution pH=7.2 in a model-independent and model-dependent way. I calculated the difference between the two models, and showed that the protein denaturation can be described as a reversible transition between two states. The results of my calculation are comparable to the results from the literature.
