Iskanje ksilanaz in celulaz v vodnih ekstraktih gliv iz slovenskih gozdov

Abstract
Lignocelulozna biomasa je osnovna strukturna komponenta rastlinske celične stene, ki vsebuje celulozo, hemicelulozo in lignin ter predstavlja vir obnovljive energije in potencialnih industrijskih surovin. Glive so učinkoviti razkrojevalci organskih snovi in polimernih spojin, saj v svoje okolje izločajo številne hidrolitične in oksidativne encime, ki so lahko tudi biotehnološko zanimivi. Glavni encimi vključeni v razgradnjo celuloze in hemiceluloze so glikozid hidrolaze, kamor uvrščamo tudi celulaze in ksilanaze. Cilji magistrske naloge so bili i) pripraviti vodne ekstrakte iz 28 gliv, ki so bile nabrane v slovenskih gozdovih, ii) ugotoviti, ali vodni ekstrakti nabranih gliv vsebujejo encime, ki sodelujejo pri razgradnji celuloze in hemiceluloze ter iii) ugotoviti, v katerem območju pH imajo optimalno delovanje. V določenih glivnih ekstraktih smo odkrili encimske aktivnosti, ki ustrezajo celulazam in ksilanazam. Med njimi smo identificirali tudi take, pri katerih celulazne in ksilanazne aktivnosti doslej še niso bile opisane v literaturi. Ugotovili smo, da imajo celulaze in ksilanaze optimum delovanja pri kisli vrednosti pH.

Title:Screening for xylanases and cellulases in crude water extracts of fungi harvested from Slovenian woods
Abstract:
Lignocellulosic biomass is the basic structural component of the plant cell wall, containing cellulose, hemicellulose and lignin, and is a source of renewable energy and potential industrial raw materials. Fungi are effective decomposers of organic matter and polymeric compounds as they secrete numerous hydrolytic and oxidative enzymes, including those of biotechnological interest. The most important enzymes involved in the degradation of cellulose and hemicellulose are glycoside hydrolases, including cellulases and xylanases. The objectives of the Master's thesis were i) to prepare aqueous extracts from 28 fungi collected in Slovenian forests, ii) to determine whether fungal aqueous extracts contain enzymes involved in the degradation of cellulose and hemicellulose, and iii) to determine the pH range for their optimal activity. In certain fungal extracts, we were able to detect enzymatic activities corresponding to cellulases and xylanases. Among them, we identified some species whose cellulase and xylanase activities have not yet been described in the literature. We have found that cellulases and xylanases have optimal activity in the acidic pH range.
