Abstract
Zaradi omejitev oralnega vnašanja učinkovin v človeško telo, se iščejo novi bolj učinkoviti načini tarčnega vnašanja učinkovin. Raziskave kažejo, da so hidrogeli, zaradi njihove podobnosti človeškim tkivom in zmožnosti počasnega sproščanja učinkovin, potencialen material na tem področju. V raziskavi smo preučevali vplive različnih parametrov na hitrost sproščanja učinkovine iz hidrogelov na osnovi alginata in TEMPO modificirane nanoceluloze. Ta dva hidrogela smo zbrali zaradi njunih strukturnih in reoloških lastnosti, kot tudi zaradi njune biokompatibilnosti, netoksičnosti in enostavne priprave. Kot glavni dejavnik, ki vpliva na hitrost sproščanja, smo opazovali vpliv stopnje zamreženja hidrogela. Prav tako pa smo opazovali hitrost sproščanja v odvisnosti od ionske moči medija sproščanja in temperature medija. Spremljali smo tudi vpliv, ki ga ima različno nabita površina hidrogela in učinkovine na sproščanje. Glede na rezultate smo določili kinetične parametre modela za napovedovanje hitrosti sproščanja iz podobnih sistemov.

Title:Controlled release of proteins from hydrogels based on modified nanocellulose and alginate
Abstract:
Because of limitations of oral drug delivery into the human body, there is a great interest in development of new drug delivery systems. Hydrogels show great potential in this field because of their similarity with human tissue and their ability to slowly release drug into surrounding medium. In our research we studied effect of different parameters on release rate of proteins from alginate and TEMPO oxidized nanocellulose hydrogels. Those two were chosen because of their structural and rheological properties as well as their biocompatibility, non toxicity and ease of preparation. We mostly observed the effect of crosslinking density on release rate, as well as effects of ionic power of release medium and temperature of release medium. We also observed if difference in surface charge between hydrogel and protein surface would affect the release rate. Based on our result we developed simple empirical model for predicting release rate from similar systems.
