Abstract
Žita in psevdožita so pomemben vir makro in mikroelementov. Zaradi fizioloških, kemijskih in bioloških lastnosti tal ter antropogenih aktivnosti se pojavlja pomanjkanje mikroelementov v rastlinah in posledično v prehrani ljudi. Z učinkovito biofortifikacijo rastlin bi rešili problem mineralne podhranjenosti. V nalogi smo preučili vpliv hladne kisikove plazme in namakanja zrn v raztopinah obogatenih s selenom oziroma cinkom, na kaljivost in mineralno sestavo kalic tatarske ajde (Fagopyrum tataricum (L.) Gaertn.) in navadne ajde (Fagopryum esculentum Moench). Za biofortifikacijo kalic s selenom smo uporabili kalijev selenat, za biofortifikacijo kalic s cinkom pa cinkov klorid. Z namakanjem zrn v raztopinah selena in cinka nam je uspelo biofortificirati kalice tatarske in navadne ajde, pri čemer vpliva na kalitev ni bilo. Analizo mineralne sestave kalic smo izvedli z rentgenskim fluorescenčnim spektrometrom in s sistemom masne spektrometrije z induktivno sklopljeno plazmo. Biofortificirane kalice tatarske ajde so vsebovale do 0,184 mg kg- 1 selena v suhi snovi (ss) in do 280 mg kg-1 cinka v ss. Biofortificirane kalice navadne ajde pa so vsebovale do 0,255 mg kg-1 selena v ss in do 323 mg kg-1 cinka v ss. Za obdelavo zrn tatarske in navadne ajde s hladno plazmo smo uporabili radiofrekvenčno kisikovo plazmo. Obdelava s hladno kisikovo plazmo se ni dokazala za učinkovito pri biofortifikaciji kalčkov ajde. Raziskava je dokazala učinkovitost namakanja zrn v mineralno obogatenih raztopinah za povečanje koncentracije Se in Zn v kalčkih tatarske in navadne ajde.

Title:Effects of biofortification of plasma-treated buckwheat (Fagopyrum sp.) grain via grain priming with zinc and selenium on germination and mineral composition of sprouts
Abstract:
Cereal and pseudocereal grains are an important source of macro and microelements. Because of physiological, chemical and biological characteristics of soil and anthropogenic activities the lack of microelements is common in plants and therefore in human nutrition. Effective microelement biofortification of plants could mitigate our mineral malnutrition. Effects of grain priming with selenium (potassium selenate was used) and zinc (zinc chloride was used) and cold radiofrequency oxygen plasma treatment on germination and mineral composition of buckwheat sprouts were studied. Two buckwheat species were investigated, namely Tartary buckwheat (Fagopyrum tataricum (L.) Gaertn.) and common buckwheat (Fagopryum esculentum Moench). Grain element composition was determined using X-ray fluorescence spectrometry and inductively coupled plasma mass spectrometry. Grain priming biofortified buckwheat sprouts without affecting the grain germination. Selenium concentration in biofortified Tartary buckwheat sprouts was up to 0,184 mg kg-1 dry weight (dw) and zinc concentration reached up to 280 mg kg- 1 dw. The highest selenium concentration in biofortified common buckwheat sprouts was 0,255 mg kg- 1 dw and of zinc up to 323 mg kg-1 dw. Plasma treatment did not improve biofortification of buckwheat sprouts. Our results confirm that grain priming with selenium and zinc rich solutions is an effective way to increase selenium and zinc concentrations in common and Tartary buckwheat sprouts.
