Razporeditev mehanoreceptorjev PIEZO1 in PIEZO2 v uroteliju sečnega mehurja

Abstract
Zaznavanje mehanskih dražljajev, ki jih povzroča raztezanje sečnega mehurja ob polnjenju z urinom, je pomembno za pravilno delovanje sečil med miksijskimi ciklusi. Pri tem so ključni mehanoreceptorji v uroteliju sečnega mehurja. Nedavne raziskave so pokazale, da celice urotelija izražajo številne mehanoreceptorje, med njimi tudi ionska kanala PIEZO1 in PIEZO2. Njuna razporeditev v uroteliju pa do danes še ni bila pokazana. Izražanje PIEZO1 in PIEZO2 v steni sečnega mehurja smo preverjali s prenosom western, njuno lokalizacijo v različnih skladih urotelija in različnih področjih mišjega sečnega mehurja pa smo proučevali z imunofluorescenco. Izbiro protiteles, ki prepoznavajo različne epitope proteinov PIEZO, smo opravili po analizi aminokislinskih zaporedij. Izražanje PIEZO1 smo dokazali z enim od treh preizkušanih protiteles, medtem ko nobeno protitelo proti PIEZO2 ni pokazalo lis na ustrezni molekulsku masi. Na zamrznjenih rezinah so bile reakcije za PIEZO1 pozitivne v bazalnih, intermediarnih in površinskih dežnikastih celicah z dvema protitelesoma. V dežnikastih celicah je bila reakcija prisotna na apikalni plazmalemi. Podobno razporeditev kaže tudi eno protitelo proti PIEZO2, pri čimer značilne označenosti apikalne plazmaleme ni mogoče zaznati. Razlik v razporeditvi obeh PIEZO nismo opazili niti na rezinah iz različnih predelov sečnega mehurja, niti pri analizi urotelija s konfokalnim mikroskopom. Z uporabo več protiteles in različnih imunohistokemijskih metod smo dokazali prisotnost mehanoreceptorjev PIEZO v bazalnem, vmesnem in površinskem skladu urotelija vseh predelov normalnega sečnega mehurja miši. S tem smo postavili temelje za študij mehanosenzorne vloge proteinov PIEZO pri boleznih sečnega mehurja, ki povzročajo nepravilnosti v mikcijskem ciklusu.

Title:Distribution of mechanoreceptors PIEZO1 and PIEZO2 in the urothelium of the urinary bladder
Abstract:
Mechanosensation induced by urinary bladder distensions in response to bladder filling is critical for proper urinary tract function during micturition cycles. Mechanoreceptors in the bladder urothelium play a crucial role. Recent studies have shown that urothelial cells express numerous mechanoreceptors, including ion channels PIEZO1 and PIEZO2, but their distribution in urothelium has not been confirmed. We investigated PIEZO1 and PIEZO2 expression with western blotting and their localization in various urothelial layers and regions of mouse urinary bladder with immunofluorescence. Selection of antibodies, which recognize different epitopes of both PIEZO proteins, was performed after amino acid sequence analysis. We confirmed PIEZO1 expression with one out of three antibodies while neither of the two PIEZO2 antibodies showed appropriate bands by western blottings. Two PIEZO1 antibodies resulted in positive labeling of basal, intermediate and superficial umbrella cells on cryosections. In umbrella cells reaction was confined to their apical plasma membrane. Similar distribution was observed by one PIEZO2 antibody, except that the apical plasma membrane was not distinctly labeled. No differences were detected between various regions of the urinal bladder urothelium in cryosections and in en face preparations for confocal microscope. Using a variety of antibodies and immunohistochemical methods we have shown PIEZO mechanoreceptors in basal, intermediate and superficial layer of the urothelium in all regions of the mouse urinal bladder. We have established a foundation for further studies of mechanosensory function of the PIEZO proteins in urinary tract diseases that affect normal micturition.
