Abstract
Virus rjave grbančavosti plodov paradižnika (ToBRFV) je bil prvič odkrit leta 2014 v Izraelu in od takrat globalno povzroča gospodarsko škodo in ogroža pridelavo paradižnika in paprike. Najpomembnejši način vnosa v nove države ali regije je preko okuženega semenskega in sadilnega materiala. Namen naloge je bil ugotoviti ali lahko pri hidroponskem načinu gojenja paradižnika pride do sproščanja virusa ToBRFV iz okuženih rastlin preko korenin v hranilno raztopino in določiti največjo razredčino tega virusa v vodi, ki jo še lahko zaznamo, saj je to poznavanje nujno za nadaljnje študije proučevanja možnosti prenosa virusa ToBRFV preko vodnih medijev. S poskusi gojenja rastlin v karantenskem rastlinjaku smo ugotovili, da se iz korenin okuženih rastlin v hranilno raztopino lahko izločajo infektivni delci virusa ToBRFV. Z analizo razredčin virusa ToBRFV v vodi pa smo ugotovili, da za uspešno mehansko okužbo testnih rastlin in za zaznavo s hitrimi serološkimi testi potrebujemo več virusnih delcev kot jih potrebujemo za njegovo zaznavo z molekularnim testom obratnega prepisovanja in verižne reakcije s polimerazo v realnem času.

Title:Tomato brown rugose fruit virus - the possibility of water-mediated transmission
Abstract:
Tomato brown rugose fruit virus (ToBRFV) was first detected in Israel in 2014 and has since caused economic damage and threats to tomato and pepper production worldwide. The main route of introduction into new countries or regions is through infected seeds and seedlings. The objective of this work was to determine if ToBRFV can be released from infected plants through the roots into the nutrient solution when plants are grown in hydroponics, and to determine the maximum dilution of this virus in water that can still be detected. This knowledge is essential for further studies on the possible transmission of ToBRFV through aquatic media. During the experiment in a quarantine greenhouse, we found that infectious particles of ToBRFV can be released from the roots of infected plants into the nutrient solution. By analyzing dilutions of ToBRFV in water, we found that we needed more virus particles for successful mechanical inoculation of test plants and for detection with rapid serological tests than for detection with a molecular assay - reverse transcription real-time polymerase chain reaction.
