Abstract
Pred masnospektrometrično analizo je N-glikane potrebno sprostiti iz glikoproteinov, nato pogosto sledi derivatizacija N-glikanov za izboljšanje občutljivosti masne spektrometrije, učinkovitejšo ionizacijo spojin in zaščito funkcionalnih skupin. Ker se z masno spektrometrijo ne da razlikovati med vsemi izomeri, se te loči s sklopitvijo s separacijsko tehniko. Najpogosteje se za ionizacijo uporabljata laserska desorpcija s pomočjo matrice in elektrorazprševanje, ki povzročita malo ali nič fragmentacije. Iz masnega spektra se tako lahko določi molekulska masa analita. Informacijo o strukturi N glikana se dobi iz tandemskega masnega spektra na podlagi ionskih fragmentov. Z visokoločljivostno masno spektrometrijo je možno razlikovati med izobarnimi spojinami. N-glikane se lahko detektira tudi z drugimi tehnikami, vendar le masna spektrometrija poda točno molekulsko maso in informacijo o strukturi analita.

Title:Determination of N-glycans by mass spectrometry
Abstract:
Before mass spectrometry analysis, N-glycans must be released from glycoproteins, usually followed by N-glycan derivatization. Derivatization improves sensitivity in mass spectrometry, ionization efficiency and protects functional groups. A lot of isomers cannot be distinguished by mass spectrometry analysis, therefore coupling with separation technique is needed to separate isomers. Matrix-assisted laser desorption/ionization and electrospray ionization are most common ionization modes, which cause little fragmentation or none at all. Molecular mass of analyte can be determined from mass spectrum. N-glycan structural information can be provided by ion fragments in tandem mass spectrum. Isobaric compounds can be differentiated by high-resolution mass spectrometry. N-glycans can be detected by other detection techniques as well, but accurate molecular mass and structural information can be provided by mass spectrometry only.
