Abstract
Namen naše raziskave je bil ugotoviti, ali je koncentracija plazme zunajceličnih veziklov pri aktivni diabetični Charcotovi nevroartropatiji (CNA) povezana z vnetnimi označevalci, zvišano temperaturo v prizadetem stopalu in koncentracijo topnega receptorja za napredne končne produkte glikacije (RAGE). Zunajcelične vezikle smo izolirali iz periferne krvi 36 bolnikov z aktivno CNA in 25 zdravih kontrolnih oseb, nato so bili na podlagi pretočne citometrije prešteti po večkratnem centrifugiranju in izpiranju vzorcev. Temperatura stopala je bila izmerjena z infrardečim termometrom. Koncentracija topnega receptorja za napredne končne produkte glikacije (RAGE) je bila ugotovljena z encimsko imunoadsorpcijsko preiskavo (ELISA). Ugotovljene so bile statistično pomembne korelacije koncentracij zunajceličnih veziklov (ne pa tudi koncentracije topnega RAGE) s C-reaktivnim proteinom (CRP) in s temperaturnimi razlikami med prizadetim in kontralateralnim stopalom (r = 0,40, p = 0,032 oziroma r = 0,84, p < 10-8). Potrjeno je bilo, da je koncentracija zunajceličnih veziklov povezana s povišano vrednostjo vnetnih označevalcev (tj. CRP in razlike v temperaturi obeh stopal) pri preiskovancih z aktivno diabetično CNA. Zunajcelične vezikle bi bilo v prihodnje smiseln obravnavati kot potencialne označevalce za postavitev diagnoze aktivne CNA, kar bi zagotovo pripomoglo k zgodnji postavitvi diagnoze in zdravljenju bolezni.

Microvesiculation of blood cells in patients with Charcot Neuroarthropathy secondary to diabetes mellitus : doctoral dissertation
Abstract:
This study focuses on membrane microvesiculation and its possible connection to inflammation processes in diabetic Charcot Neuroarthropathy (CNA). It was our aim to determine whether the concentration of extracellular vesicles (EVs) in isolates from blood of patients with CNA is increased, and whether it is connected to the concentration of soluble receptor for advanced glycation end products (RAGE) and standard markers of inflammation. EVs were isolated from peripheral blood of 36 patients with active CNA and of 25 non-Diabetic control subjects.
