Abstract
Uporaba ogrodnih sistemov kot nosilcev prirejenega sproščanja je zaradi številnih prednosti v porastu. V ta namen se najpogosteje uporablja hipromeloza (HPMC), ki ima ob stiku z vodnimi raztopinami visoko sposobnost tvorbe gela. V okviru magistrske naloge smo pripravili ogrodne tablete s hipromelozo in karvedilolom kot modelno zdravilno učinkovino. Z valjčnim kompaktiranjem smo iz praškaste zmesi pripravili kompaktate sestavljene iz različnih polnil. Kompaktate smo po izdelavi mleli z različnimi vrstami mlinov pri izbranih procesnih parametrih mletja, nastale granule pa tabletirali v ogrodne tablete. Preveriti smo želeli, ali in kako razlike v vrsti uporabljenega polnila ter načinu mletja kompaktatov vplivajo na fizikalne in partikulatne lastnosti granul in sproščanje zdravilne učinkovine iz končne farmacevtske oblike. Na velikost granul najbolj vpliva izbira mlina za mletje, poleg tega pa je velikost granul odvisna tudi od velikosti odprtin uporabljene mreže. Nakazuje se tudi vpliv vrste uporabljenega polnila. Na sproščanje karvedilola iz ogrodnih tablet s hipromelozo vplivata vrsta uporabljenega mlina ter uporabljeno polnilo, pri čemer je vpliv vrste uporabljenega mlina preko vpliva na velikost granul izrazitejši v začetnem delu profila sproščanja, po vzpostavitvi gelskega plašča pa začne prevladovati vpliv uporabljenega polnila.

Title:Effects of ribbons composition and milling on the release of carvedilol from hypromellose-based matrix tablets
Abstract:
The use of matrices as modified-release carriers is increasing due to its many advantages. Hypromellose (HPMC), which has a high gel-forming ability upon contact with the aquoeus solutions, is most commonly used for this purpose. As part of the master's thesis, hypromellose matrix tablets were prepared with carvedilol as a model active ingredient. Using roller compaction, ribbons consisting of various fillers were prepared from the powdered mixture. The ribbons were subjected to milling using different types of mills by applying selected milling process parameters, and the resulting granules were formed into matrix tablets. We wanted to verify if and how the differences in the type of filler used and the method of ribbon milling affect the physical and particulate matter properties of the granules and the release of the active ingredient from the final dosage form. The size of the granules depends mostly on type of the mill. Furthermore, the size of the granules also depends on the screen size of the sieve used. The influence of the type of filler used was also indicated. The release of carvedilol from the hypromellose matrix tablets is influenced by the type of mill and the filler used, whereby the impact of the type of mill used is more pronounced in the initial part of the release profile, whereas after the gel layer has been formed, the impact of the filler used becomes more dominant.
