Vpliv in privzem izbranih nanomaterialov pri praživali Tetrahymena thermophila in celični liniji SH-SY5Y

Abstract
Namen magistrskega dela je bil preveriti celični privzem in učinke različnih nanomaterialov na dva tipa celic: humano celično linijo SH-SY5Y in pražival Tetrahymena thermophila. Preverjali smo tudi, kako predhodno ustvarjena proteinska korona iz govejega serumskega albumina (BSA) vpliva na učinek nanodelcev. Za testiranje smo uporabili tri vrste nanodelcev TiO2 in eno vrsto CuO v koncentracijah od 10 do 75 μg/mL. Po izpostavitvi celic SH-SY5Y nanodelcem smo uporabili tri teste za zaznavanja citotoksičnosti: test z resazurinom, test privzema barvila nevtralno rdeče in test Coomassie brilijant modro. Za preverjanje toksičnosti nanodelcev na T. thermophila smo uporabili samo test z resazurinom. Za preverjanje privzema delcev v celice, smo pri celični liniji SH-SY5Y uporabili test privzema barvila nevtralno rdeče, pri T. thermophila pa svetlobno mikroskopijo. Ugotovili smo, da je celična linija SH-SY5Y bolj občutljiva na izpostavitev delcem kot T. thermophila. Na SH-SY5Y so citotoksično delovali nanodelci CuO in v manjši meri ena vrsta nanodelcev TiO2. Predhodno ustvarjena proteinska korona je v večini primerov zmanjšala kvarni vpliv TiO2 in CuO nanodelcev na celice SH-SY5Y, na pražival Tetrahymena thermophila pa ni imela vpliva. Potrdili smo privzem nanodelcev v celice Tetrahymena thermophila, ne pa tudi v celice SH-SY5Y.

Title:Effect and uptake of selected nanomaterials in a protozoan Tetrahymena thermophila and cell line SH-SY5Y
Abstract:
The intention of master thesis' work was to test the cellular uptake and effects of various nanomaterials on two types of cells: human cell line SH-SY5Y and protozoan Tetrahymena thermophila. We were checking whether nanoparticles with bovine serum albumin (BSA) protein corona affects possible cytotoxicity of nanoparticles. We chose three types of TiO2 and one type of CuO nanoparticles at concentrations from 10 to 75 μg/mL. After treating SH-SY5Y with nanoparticles we used three tests to determine cytotoxicity: resazurin assay, neutral red assay and Coomassie brilliant blue assay. In case of Tetrahymena thermophila we used only resazurin assay. To determine cellular uptake of nanoparticles in SH-SY5Y cells neutral red assay was used, whereas light microscopy for Tetrahymena thermophila. Our results revealed that cell line SH-SY5Y is more sensitive to the effects of nanoparticles than T. thermophila. CuO and one type of TiO2 nanoparticles displayed some cytotoxic effects on SH-SY5Y. In most cases protein corona diminished the effect of TiO2 and CuO nanoparticles on SH-SY5Y but did not affect the toxicity on Tetrahymena thermophila. We confirmed cellular uptake of nanoparticles in Tetrahymena thermophila but not in SH-SY5Y.
