Abstract
Žlahtna vinska trta (Vitis vinifera L.) je zaradi svoje ekonomske pomembnosti ena najbolj razširjenih kmetijskih rastlin. Pridelovalci stremijo k čim večjemu in kakovostnejšemu pridelku, poslužujejo se hitrega vegetativnega razmnoževanja, ki predstavlja tveganje za pojav in širitev virusnih obolenj. Rešitev nudi in vitro vzgoja brezvirusnega sadilnega materiala s kulturo meristemov in somatsko embriogenezo v kombinaciji s termoterapijo in kemoterapijo, če je potrebno. Rastlinsko tkivo je zaradi svojega totipotentnega potenciala zelo primerno in nudi dobro diferenciacijo ter regeneracijo izoliranega meristemskega tkiva. Za določitev oz. identifikacijo virusov v rastlinskem tkivu se poslužujemo serološkega ELISA testa, ki na osnovi protiteles in antigenov v rastlinskem soku ovrže ali potrdi prisotnost virusa. ELISA test je eden najpogosteje uporabljenih testov za določanje virusov, saj je enostaven in dobro rutiniran z možnostjo uporabe že vnaprej pripravljenih setov. Molekulska PCR metoda predstavlja natančnejšo metodo identifikacije virusov, kjer lahko z načrtno oblikovanimi začetnimi oligonukleotidi ali večstopenjskimi izvedbami določimo prisotnost večih virusov. Diplomska naloga nudi pregled omenjenih tehnik in metod ter njihove prednosti oz. slabosti.

Title:Production of virus-free grapevine Vitis vinifera L. planting material in vitro.
Abstract:
Grapevine (Vitis vinifera L.) is due to its economic importance one of the most common agricultural crops. Manufacturers aim to large and qualitative harvest, they tend to fast vegetative reproduction, which often leads to the transmission of viruses. The key in preventing so can be in vitro virus-free production of planting material such as meristem culture and somatic ambryogenesis in combination with thermotherapy and chemotherapy if needed. Plant tissue is due to its totipotent potential very appropriate and offers good diferentiation and regeneration of isolated meristem tissue. For the determination and identification of the viruses in plant material serological ELISA test can be used based on the antibody and the antigen complex, which refutes or confirms the presence of the virus. ELISA is due to its simplicity and routine kits, which ca be prepared in advance, one of the mostly used tests for virus determination. Molecular PCR method presents more accurate method for virus identification, where specially designed primers can be used or method itself can be performed in more than just one stage, enabling us identification of several viruses.
