Abstract
RNA-vezavne proteine se običajno obravnava kot regulatorje izražanja genov na post-transkripcijskem nivoju. Njihova vezava poteka preko ene ali več domen in vpliva na usodo ter funkcijo molekule RNA. Odkritih je bilo več sto takšnih proteinov z dobro poznanimi domenami. Nedavno so študije na nivoju celotnega intraktoma prispevale k odkritju velikega števila popolnoma neraziskanih RNA-vezavnih proteinov z novimi načini vezave in delovanja. Izjemno pomembno vlogo pri preučevanju medsebojnega delovanja igrajo najsodobnejše metode za zajetje interaktoma in študije vezave v živih celicah. Poleg tega so se zaradi izjemne količine podatkov uveljavili računalniški pristopi, ki lahko bistveno olajšajo odkrivanje novih načinov vezave kot tudi pojasnjevanje obstoječih. V svojem delu obravnavam primere neobičajnih načinov vezave in najmodernejše pristope za vpogled v svet RNA-vezavnih proteinov.

Title:Discovering new RNA-binding proteins
Abstract:
RNA-binding proteins are usually thought of as regulators of gene expression at a post-transcriptional level. By binding with multiple domains, they modulate the faith and function of RNA molecules. Throughout the years several hundreds of them were found to bind RNA with well-known domains. Recently interactome-wide studies have contributed to the discovery of several completely unknown RNA-binding proteins with new modes of binding and function. Crucial for their discovery were modern methods for RNA interactome capture and analysis of RNA-protein interactions from a living cell. Furthermore, enormous amounts of data enabled emergence of computational approaches that facilitate the discovery of new modes of binding as well as clarification of the existing ones. In my work I illustrate unconventional ways of binding and expound state-of-the-art methods for the analysis of the RNA-binding proteins.
