Abstract
Bakterije iz rodu Stretomyces proizvajajo veliko različnih bioloških učinkovin, med katere spadajo različni tetraciklinski antibiotiki, kot na primer oksitetraciklin. Tetraciklini so antibiotiki širokega spektra. Za industrijsko produkcijo oksiteraciklina se uporabljajo netopna kopleksna gojišča, kar velikokrat privede do težav pri analizi fermentacijskega procesa in drugih parametrov bioprocesne brozge. Cilj magistrske naloge je razvoj topnega in kemijskega definiranega gojišča za produkcijo oksitetraciklina. Netopne komponente kompleksnega gojišča smo nadomeščali s topnimi alternativami in iskali ustrezne koncentracije. CaCO3 smo zamenjali s CaCl2 in dodali pufer MOPS ali MES, kot topen vir ogljika smo uporabli dekstrin in kot topen vir dušika amonijev sulfat. Iz gojišča smo odstranili CSL in v gojišče dodali mineralno mešanico. Z nadomeščanjem komponent gojišča sicer nismo dosegali visokih donosov oksitetraciklina, vendar so bili ti donosi še vedno v sprejemljivih vrednostih. Kompomente smo nadomeščali v gojiščih vegetativne in produkcijske stopnje.

Title:Development of soluble and chemically defined medium for biosynthesis of oxytetracycline with Streptomyces rimosus.
Abstract:
Members of genus Streptomyces are know producers of various natural products, including different tetracycline antibiotics such as oxytetracycline. Tetracyclines are broad-spectrum antibiotics. Non-soluble complex media are used for industrial production of oxytetracycline; often causeing signifficant difficulties in analysis of fermentation process and other culture parameters.. It was therefore goal of this research development of soluble and chemically defined medium for biosynthesis of oxytetracycline. We replaced non-soluble components of complex medium with soluble alternaives. We replaced CaCO3 with CaCl2 with addition of buffer MOPS or MES. As soluble source of carbon we used dextrin and as soluble soruce of nitrogen we used ammonium sulphate. CSL has been removed from medium and mix of minerals was added. Athough much lower yiled of oxytetracycline was achieved, we stilč manage to retain sifficient yield by replacing different medium components in both, vegetative and productive media.
