Učinek potencialnih efektorjev fitoplazme Candidatus Phytoplasma solani na rastlinski fenotip

Abstract
Evropa je glavni svetovni proizvajalec in izvoznik sadik vinske trte (Vitis vinifera L.) in vina. Ta pomembni gospodarski odsek ogrožajo številne bolezni vinske trte, med njimi so še posebej pomembne glivne bolezni in bolezni trsnih rumenic (GY), ki jih povzročajo fitoplazme, fitopatogene bakterije iz razreda Mollicutes. V Evropi sta na vinski trti zelo razširjeni navadna trsna rumenica ali počrnelost lesa (Bois noir, BN), ki jo povzroča 'Candidatus Phytoplasma solani' (Bois Noir phytoplasma, BNp), in zlata trsna rumenica (Flavescence dorée, FD), ki jo povzroča karantenska fitoplazma FDp. S pomočjo bioinformatskih orodij smo iz kataloga potencialnih efektorjev BNp, ki ga je pripravila skupina dr. Günterja Braderja z Avstrijskega inštituta za tehnologijo, izbrali šest potencialnih efektorjev. Sekvence izbranih efektorjev smo s postopki kloniranja vstavili v binarne vektorje ter jih agroinfiltrirali v liste tobaka (Nicotiana benthamiana L.). S konfokalno mikroskopijo smo pokazali, da se je vseh šest efektorjev izražalo v jedru in citoplazmi transformiranih celic. Poskusili smo vpeljati tudi postopek za prehodno transformacijo vinske trte.

Title:The effect of potential Candidatus Phytoplasma solani effectors on the plant phenotype
Abstract:
Europe is the most important producer and exporter of vine seedlings (Vitis vinifera L.) and wine. This important economic section is endangered by many different diseases, fungi diseases and grapevine yellows (GY) diseases. Responsible for GY diseases are phytoplasmas, phytopathogenic bacteria from class Mollicutes. In Europe there are two major phytoplasma groups that cause GY. 'Candidatus Phytoplasma solani' which is associated with yellows disease ('Bois noir', BN). And second yellows disease (Flavescence dorée, FD) which is associated with quarantine phytoplasma FDp. Using bioinformatic tools, we chose from effector catalogue, which was prepared by dr. Günter Brader scientific group from Austrian institute for technology, six potencial effectors. We inserted sequences of chosen effectors in to binary vectors and agroinfiltrated them in to tobacco leaves.. Using confocal microscopy, we showed that all six effectors localized in nucleous and cytoplasma of transformed cells. We tried to introduce a grapevine Agrobacterium-mediated transient transformation protocol to our lab.
