Abstract
Revmatoidni artritis je kronična vnetna bolezen, ki povzroča postopne poškodbe sklepov in počasno izgubljanje gibalne funkcionalnosti. Področje vnetnih revmatičnih bolezni je ena najhitreje se razvijajočih medicinskih strok. Vsem tem boleznim je skupen predvsem neustrezen imunski odziv na telesu lastne antigene. Čeprav revmatoidni artritis poznamo že veliko časa in je to relativno stara bolezen, se je njeno zdravljenje in doktrina zdravljenja spremenila ter močno izboljšala šele v zadnjem desetletju, k temu pa je zelo pripomogel predvsem razvoj in napredek na področju bioloških zdravil. Boljše razumevanje patogeneze bolezni je prispevalo k boljšemu in učinkovitejšemu razvoju bioloških zdravil za zdravljenje avtoimunskih bolezni. Biološka zdravila ciljano zavirajo aktivnost imunsko kompetentnih celic oziroma njihovih proizvodov - citokinov in predstavljajo pomemben del klinične prakse pri zdravljenju težjih oblik revmatoidnega artritisa. V revmatologiji uporabljamo predvsem zaviralce TNF-α, interlevkinov 6, zaviralce CD20+ celic B in zaviralce T- celične kostimulacije. Cilj moje naloge je raziskati in podati ključne informacije o delovanju bioloških zdravil, ki so nam trenutno na voljo in so v uporabi.

Title:Molecular mechanisms of biological drugs in theraphy of rheumathoid arthritis
Abstract:
Rheumatoid arthritis is a chronic inflammatory disease that causes deformed joints and leads to gradually reduced mobility. The field of rheumatic diseases is one of the fastest developing medical disciplines. All the rheumatic diseases share an abnormal immune response to the antigens. Although rheumatoid arthritis is not a new disease, it is only in the last decade that we are facing positive changes in its medical treatment due to development of biological drugs. Better understanding of pathogenesis of the disease opened the doors for more efficient development of medication for treatment autoimmune diseases. Biological medications target and block the activity of immune competent cells and their products - cytokins and play a big role in clinical treatment of severe forms of rheumatoid arthritis. In rheumatology, we use mainly inhibitors of TNF-α and interleukins 6, inhibitors of CD20+ B cells and T-cell activation inhibitors. The objective of my thesis is to research and introduce the key information about the mechanism of action of biological medications that are currently being used.
