Abstract
S hipoksijo inducirana podenota transkripcijskega dejavnika HIF1A je ena izmed ključnih komponent signalne verige pomanjkanja kisika. Uravnava izražanje velikega števila genov, prevzema osrednjo vlogo pri uravnavanju homeostaze kisika in usklajuje izražanje genov, povezanih s fiziološkimi in patološkimi procesi prilagajanja. Z namenom pridobitve celostnega vpogleda v odziv na pomanjkanje kisika smo v diplomskem delu zbrali poznane direktne tarčne gene HIF1A, jih klasificirali glede na njihovo vključenost v bioloških poteh, naredili pregled celičnih linij, znotraj katerih je bila dokazana neposredna vezava transkripcijskega dejavnika in podatke prikazali v preglednicah. Nadalje smo temo poglobili in se osredotočili na direktne povezave, dokazane v celičnih linijah raka debelega črevesja in danke ter podrobneje analizirali šest izbranih genov; ABCB1, ADM, ANKRD37, BNIP3, CAD in COX4I2.

Title:The Targetome of Hypoxia Induced HIF1A Transcription Factor in Human
Abstract:
The hypoxia‒induced transcription factor subunite HIF1A of is one of the primary signal chain components of oxygen deficiency. It governs a large number of genes, has a central role in the regulation of oxygen homeostasis and manages the expressing of genes, connected with physiological and pathological processes of adjustment. With the purpose of gaining the necessary insight into the cellular response of oxygen deficiency, we collected the known direct target genes of HIF1A, classified them according to their involvement in various biological pathways, performed an overview of cell lines in which the direct binding of the transcription factor was proven and systematized all findings in tables. Furthermore, this work is focused on the direct links proven in the cell lines of colorectal cancer and analyzed in detail, the function and connection to HIF1A in six selected genes; ABCB1, ADM, ANKRD37, BNIP3, CAD, and COX4I2.
