Abstract
Pri metabolizmu raka običajno pride do velikih sprememb v izražanju in regulaciji proteinov. 6-fosfofrukto-1-kinaza je eden od pomembnih encimov, ki uravnavanjo tok glikolize in pri katerem pride do velikih sprememb, ki posledično vodijo v deregulacijo pretoka metabolitov. Med trenutno opaženimi spremembami fosfofruktokinaze so spremenjeno izražanje izooblik encima, pojav kratkih aktivnih fragmentov in neodzivnost na inhibicijo. Poskusili smo najti razlago, zakaj se pri raku pojavljajo kratki fragmenti, kako nastanejo in kakšne so njihove lastnosti. Opravili smo poravnavo aminokislinskih zaporedij izooblik in opazovali razlike med njimi, ter poskusili določili potencialna cepitvena mesta in na podlagi le-teh poskušali najti proteinazo, odgovorno za nastanek kratkih fragmentov.

Title:Characterization of isoenzymes 6-phosphofructo-1-kinase in tumorigenic cell lines
Abstract:
In cancer metabolism, changes in expression and regulation of protein are common. 6-phosphofructo-1-kinase is one of the important enzymes that regulates metabolic flow of glycolysis, and where it comes to drastic changes in enzyme, which consequently leads to deregulation of metabolic flow. Among currently noticed changes in photofructokinase are substitutions of isoenzyme form, creation of short, active fragments and no response to inhibition. We tried to find explanation, why do short fragments appear in case of cancer, how are they created and what features they have. We aligned the aminoacid sequences of isoenzymes in order to find potential cleaving sites and with them as the base, tried to find proteinase responsible for occurene of short fragments.
