Abstract
Histamin je biogeni amin, ki se sprošča pri vnetju iz krvnih in tkivnih bazofilcev. Povzroča vazodilatacijo in povečano prepustnost kapilar. Sproščanje histamina lahko zavirajo različne učinkovine. Med njimi so steroidne in nesteroidne protivnetne učinkovine. V nalogi smo želeli ugotoviti, kako izbrane steroidne in nesteroidne protivnetne učinkovine vplivajo na sproščanje histamina iz tkivnih bazofilcev peritonealne votline podgane, aktiviranih s spojino 48/80. Uporabili smo steroidne učinkovine kortizon, hidrokortizon in prednizolon ter nesteroidne učinkovine ketoprofen, diklofenak in indometacin. Preverili smo tudi vpliv učinkovin na sproščanje histamina iz neaktiviranih tkivnih bazofilcev. Tkivne bazofilce smo izolirali iz peritonealne votline podgane in jih izpostavili različnim koncentracijam izbranih steroidnih in nesteroidnih protivnetnih učinkovin. Sproščanje histamina iz tkivnih bazofilcev smo inducirali z uporabo neimunološkega sproščevalca, spojino 48/80. Koncentracijo histamina smo določali s spektrofluorofotometrično metodo.Ugotovili smo, da tako steroidne kot nesteroidne protivnetne učinkovine zavirajo sproščanje histamina iz tkivnih bazofilcev, sproženega s spojino 48/80. Učinek je odvisen od koncentracije uporabljene učinkovine in narašča z njo. Največji zaviralni učinek smo ugotovili pri indometacinu, ki je pri koncentraciji 10-3 mol/l skoraj v 90 % zavrl sproščanje histamina iz aktiviranih celic. Hidrokortizon je edina učinkovina, ki pri nobeni uporabljeni koncentraciji ne zavira sproščanja histamina, sproženega s spojino 48/80. Rezultati so pokazali tudi, da nobena od uporabljenih učinkovin sama po sebi ne vpliva na sproščanje histamina iz neaktiviranih tkivnih bazofilcev.

Title:Comparison of the effect on non-steroidal and steroidal agents on mast cell histamine release
Abstract:
Histamine is a biogenic amine released from blood basophils and mast cells during inflammation. It causes vasodilatation and increased capillary permeability. Histamine release could be hindered by various active substances, including steroids and non steroidal anti-inflammatory substances. In our study we wanted to determine the influence of selected steroids and non steroidal anti-inflammatory substances on histamine release from rat peritoneal mast cells activated by compound 48/80. We used cortisone, hydrocortisone and prednisolone among steroids and ketoprofen, diclofenac and indomethacin among non steroidal anti-inflammatory substrances. We also investigated the influence of the selected substances on histamine release from inactivated rat peritoneal mast cells. We isolated rat peritoneal mast cells and exposed them to different concentrations of the selected steroids and non steroidal anti-inflammatory substances. Histamine release was induced by non-immunologic secretagogue, compound 48/80. The concentration of histamine was measured spectrofluorophotometrically. While performing this study we found out that both steroids and non steroidal anti-inflammatory substances hinder histamine release from rat peritoneal mast cells activated with compound 48/80. The inhibition of histamine release depends on the concentration of the selected substance and increases with its concentration. The maximal inhibition was observed when mast cells activated by compound 48/80 were incubated with indomethacin. We noticed 90 % inhibition at a 10-3 mol/L concentration. On the other hand, hydrocortisone was the only substance with no inhibitory effect on histamine release induced by compound 48/80. In our study none of the substances had an influence on histamine release from inactivated mast cells.
