Skupne fenolne spojine in antioksidativna učinkovitost soka granatnega jabolka

Abstract
V okviru diplomskega dela smo med seboj primerjali skupne fenolne spojine in antioksidativno učinkovitost sveže iztisnjenih sokov granatnega jabolka. Ugotavljali smo razlike med ročno in strojno iztisnjenim sokom in med sokovi različnih porekel (Turčija, Indija). Pri ročni pripravi vzorca smo iztisnili samo sok iz arilov, ne da bi pri tem poškodovali seme, pri strojnem načinu pa se je poškodovalo tudi seme. Vsebnost skupnih fenolnih spojin smo v soku določili spektrofotometrično s pomočjo Folin-Ciocalteu metode, antioksidativno učinkovitost pa po metodi s prostim radikalom DPPH·. Med granatnimi jabolki turškega in indijskega porekla ni bilo signifikantne razlike v nobenem izmed analiziranih parametrov, medtem ko smo statistično značilne razlike določili v sokovih, iztisnjenimi na različna načina (ročno, strojno). Sokovi, ki so bili ročno iztisnjeni, so imeli večje vsebnosti skupnih fenolnih spojin, (6,8 ± 1,3) mM, in antioksidativno učinkovitost, (8,1 ± 2,0) mM, v primerjavi s sokovi, ki smo jih iztisnili strojno, saj so le-ti imeli vsebnost skupnih fenolnih spojin (5,7 ± 1,0) mM in antioksidativno učinkovitost (5,7 ± 1,3) mM. Dobljene vrednosti skupnih fenolnih spojin so nižje v primerjavi z raziskavo Fischerja in sod. (2011), kar je verjetno posledica razlik v zasnovi eksperimenta. Antioksidativno učinkovitost smo primerjali z raziskavami Gil in sod. (2000), Mousavinejad in sod. (2009) ter z Fischer in sod. (2011) in ugotovili, da so vrednosti precej podobne.

Title:Total phenolic compounds and antioxidant activity of pomegranate juice
Abstract:
In the course of this graduation thesis we compared total phenolic compounds and antioxidant activity in fresh squeezed pomegranates juice. We found differences between manual and machine-squeezed juice and between juices of diferent origin (Turkey, India). In the manual preparation of the sample we only pressed the juice from the arils without damaging the seeds, while in the mechanical mode, the seeds were damaged. Total phenolics were determined spectrophotometrically by means of Folin–Ciocalteu method and antioxidant activity with the free radical DPPH· assay. There was no significant difference between Turkish and Indian pomegranate in any of the analysed parameters, while the statistically significant differences were determined in differently squeezed juices (manually, machine). Manually squeezed juices had higher levels of total phenolic compounds, (6.8 ± 1.3) mM, and antioxidant efficiency, (8.1 ± 2.0) mM, compared with the machine-squeezed juices that had (5,7 ± 1,0) mM of total phenolic compounds and antioxidant efficiency (5,7 ± 1,3) mM. The obtained values of the total phenolic compounds are lower in comparison with the Fischer study et al. (2011), which can be attributed to differences in the design of the experiment. Antioxidant activities were compared to studies performed by Gil and et al. (2000), Mousavinejad et al. (2009) and by Fischer et al. (2011) quite similar.
