Abstract
Črna detelja (Trifolium pratense) je polimorfna rastlinska vrsta iz družine Fabaceae. Največ črne detelje se pridela za krmo živali, saj izboljšuje krmno vrednost, v poljedelstvu pa je pomembna zaradi sposobnosti simbiotske fiksacije dušika z Rhizobium sp. V Slovenski rastlinski genski banki (SRGB) hranijo 114 genskih virov črne detelje. Namen magistrskega dela je ocena genetske in morfološke raznolikosti slovenskih genskih virov črne detelje iz SRGB in izbranih sort, vpeljava metode določitve vsebnosti izoflavonov s tekočinsko kromatografijo visoke ločljivosti sklopljeno z masno spektrometrijo (HPLC-MS/MS) in določitev vsebnosti izoflavonov v genskih virih. Z mikrosatelitnimi markerji smo ocenili veliko raznolikost znotraj genskih virov (86,6 %), manjši delež raznolikosti pa je med posameznimi genskimi viri (13,4 %). Ovrednotili smo 15 morfoloških znakov ter genske vire razvrstili glede na morfološko različnost. Največjo vsebnost izoflavonov smo pri sorti 'Poljanka' določili s sušenjem pri 105 °C 20 minut in nadaljnjih 65 °C 5 ur v fenofazi med cvetenjem. Pri proučevanih genskih virih se je vsebnost genisteina, prunetina in ononina razlikovala. Informacija o genetski in morfološki raznolikosti ter vsebnosti izoflavonov v slovenskih genskih virih in sortah bo uporabna pri žlahtnjenju novih sort črne detelje.

Title:GENETIC AND MORPHOLOGICAL DIVERSITY OF RED CLOVER (Trifolium pratense L.) GENETIC RESOURCES FROM SLOVENIA AND ISOFLAVONE CONTENT IN SELECTED GENOTYPES
Abstract:
Red clover (Trifolium pratense) is a highly polymorphic cross-pollinating legume species from Fabaceae family. It is important as fodder for livestock, as it improves nutritional value of feed. Red clover is important in agriculture, especially because it is able to perform symbiotic nitrogen fixation by Rhizobium sp. Slovenian plant gene bank (SPGB) stores 114 red clover accessions. The aim of the work was to determine genetic and morphological diversity of red clover accessions from SPGB and selected varieties and to determine concentration and distribution of red clover isoflavones by high pressure liquid chromatography and mass spectrometry. Using microsatellite markers, high diversity within individual genetic resources (86,6 %) was detected, while diversity between different genetic resources (13,4 %) was small. Fifteen morphological traits were evaluated. Differences of isoflavone concentration and differences between different preservation methods after harvest were observed. Total isoflavone concentrations for ‘Poljanka’ variety were the highest when drying at 105 °C for 20 minutes and at 65 °C for 5 hours in flowering phenophase. The content of genistein, prunetin and ononin varied significantly between genotypes. Information on genetic and morphological diversity and content of isoflavones in Slovenian genetic resources is important for further breeding of new red clover varieties.
