Abstract
Uvod: Učinkovito pakiranje hrane je ključno za podaljšanje roka uporabnosti in zagotavljanje varnosti živil. Hitro pokvarljivo sadje, kot so hruške, je še posebej dovzetno za okužbe s plesnijo P. expansum, kar lahko privede do nastanka strupenega toksina patulina. Uporaba kvartarnega hitozana, kemično modificiranega derivata hitozana, povečuje topnost in antimikrobne lastnosti, kar znatno izboljša obstojnost in učinkovitost zaščite živil pred mikrobiološkimi okužbami. Namen: Namen magistrskega dela je raziskati, kako obdelava različnih sort hrušk s premazom iz kvartarnega hitozana vpliva na rast plesni P. expansum in zmanjšuje njene poškodbe. Metode dela: V raziskavi smo uporabili pet sort hrušk (Conference, Celina, Abate Fetel, Packham's Triumph in Williams), skupaj 99 plodov. Na očiščene hruške smo nanesli dve koncentraciji kvartarnega hitozana (0,25 % in 1 %), raztopljenega v 1 % ocetni kislini, ter jih inokulirali s plesnijo P. expansum. Rast plesni smo spremljali 14 dni, merili premer lezij, nato pa rezultate statistično obdelali ter določili vpliv vrste hrušk, koncentracije hitozana in časa inkubacije na rast plesni. Rezultati: Kvartarni hitozan zavira rast plesni P. expansum na testiranih hruškah, pri čemer različne sorte hrušk izkazujejo različno dovzetnost. Koncentracija 0,25 % kvartarnega hitozana je statistično značilno zmanjšala rast plesni pri sortah Conference in Abate Fetel. Z 1 % premazom se je uspešnost zaščite pokazala pri istih dveh vrstah hrušk, medtem, ko smo učinkovitost 1 % raztopine zaznali le okimetrično pri sorti Packham's Triumph. Razprava in zaključek: Na podlagi analize rezultatov lahko potrdimo učinkovitost kvartarnega hitozana pri zaviranju rasti plesni P. expansum le pri dveh sortah hrušk Conference in Abate Fetel, medtem ko je bil učinek pri drugih sortah manj izrazit ali neučinkovit. Razlike v biofizikalni odpornosti posameznih sort hrušk so se izkazale za ključni parameter pri izbiri ustrezne zaščite sadja.

Title:The effect of quaternary chitosan on the growth and development of Penicillium expansum on different pear cultivars after harvest : master thesis
Abstract:
Introduction: Efficient food packaging is essential to extend shelf life and ensure food safety. Perishable fruits such as pears are particularly susceptible to P. expansum mould infections, which can lead to the formation of the toxin patulin. The use of quaternary chitosan, a chemically modified derivative of chitosan, increases solubility and antimicrobial properties, which significantly improves the durability and effectiveness of food protection against microbial contamination. Purpose: it was to investigate how the treatment of different pear cultivars with a quaternary chitosan coating affects the growth of P. expansum and reduces its damage. Methods: Two concentrations of quaternary chitosan (0.25% and 1%) dissolved in 1% acetic acid were applied to cleaned pears and inoculated with P. expansum. The growth of the mould was monitored for 14 days, the diameter of the lesions was measured, and the results were statistically analysed to assess the influence of pear species, chitosan concentration and incubation time. Results: Quaternary chitosan inhibits the growth of P. expansum on the pears tested, with different pear varieties showing different susceptibility. A concentration of 0,25% quaternary chitosan reduced mould growth in Conference and Abate Fetel varieties to a statistically significant level. With the 1% coating, the protection effectiveness was observed in the same two pear varieties, while the effectiveness of the 1% solution was only detected visually in the Packham's Triumph variety. Discussion and conclusion: Based on the results, we can confirm the effectiveness of quaternary chitosan in inhibiting the growth of the mold P. expansum only in two pear varieties, Conference and Abate Fetel, while the effect in other varieties was less pronounced or ineffective. Differences in the biophysical resistance of individual pear varieties proved to be a key parameter in selecting appropriate fruit protection.
