Abstract
Diplomsko delo raziskuje vpliv pufrov in aditivov na retencijski čas proteinov v ionsko izmenjevalni kromatografiji (IEX). IEX je široko uporabljena tehnika za ločevanje proteinov na podlagi njihovih površinskih nabojev, na katere vpliva predvsem pH. Pufri igrajo ključno vlogo pri vzdrževanju pH vrednosti in zagotavljajo ponovljivost pri ločevanju proteinov. Izbira pufrov in njihova ionska moč morata biti usklajena s stabilnostjo in aktivnostjo proteinov. Aditivi, kot so soli in organska topila, vplivajo na interakcije proteinov s stacionarno fazo in vplivajo na retencijske čase s specifičnimi in nespecifičnimi učinki. Raziskava poudarja pomen optimizacije teh parametrov za doseganje učinkovitega čiščenja proteinov.

Title:Effect of buffer and additives on protein retention in ultra high pressure ion exchange liquid chromatography
Abstract:
This paper investigates the influence of buffers and additives on the retention time of proteins in ion exchange chromatography (IEX). IEX is a widely used technique for the separation of proteins based on their surface charges, which influence pH. Buffers play a key role in maintaining the final pH conditions and ensuring the reproducibility of protein separation. The choice of buffers and their ionic strength must be matched to the stability and activity of the proteins. Additives, e.g. in the organic solvent, influence the interaction of the proteins with the stationary phase and have specific and non-specific effects on the retention times. Research shows how important the optimisation of these parameters is for efficient protein purification.
