Abstract
Namen magistrskega dela je bil preučiti vpliv krme, linije in spola na navidezno prebavljivost surovih beljakovin in prebavljivost surovih maščob pri linijah FHI (vitka linija miši) in FLI (debela linija miši). Testirali smo dve vrsti krme, HFD (krma z veliko vsebnostjo maščob), LFD (krma z majhno vsebnostjo maščob), ločeno po spolu in liniji. Prebavljivostni poskus smo izvajali v 2 ponovitvah, in sicer, ko so bile živali stare 6 tednov (1ObPr) ter z odraslimi živalmi pri 13 tednih (2ObPr). Ugotovili smo, da ima večji vpliv na parametre prebavljivosti krma HFD. Ob zaključku postopka so imeli samci večjo telesno maso kot samice. Skupina samcev HFD-FHI je zaužila največ surovih beljakovin in surovih maščob, medtem ko je pri samicah največ surovih beljakovin zaužila skupina HFD-FHI surovih maščob pa skupina HFD-FLI. Boljšo navidezno prebavljivost surovih beljakovin smo izračunali pri samicah HFD-FLI in HFD-FHI v primerjavi z drugimi skupinami. Do podobnih ugotovitev smo prišli tudi pri prebavljivosti surovih maščob, saj so samice HFD-FHI in HFD-FLI imele boljšo prebavljivost surovih maščob v primerjavi s samicami na LFD ter samci obeh linij ne glede na vrsto krme. Naši rezultati so potrdili nekatere prejšnje študije na tem modelu miši, da fenotipskih razlik v prirastu in deležu telesnega maščevja med debelo in vitko linijo ne moremo pojasniti z razlikami v zauživanju krme. Večinoma v tem parametru razlik ni bilo, pri samcih pa je vitka linija zaužila celo statistično značilno več krme kot debela linija. Dokazali smo določene razlike med linijama in vpliva sestave krme pri navidezni prebavljivosti, kar bi bilo potrebno upoštevati pri nadaljnji evalvaciji fenotipskih razlik med genotipoma.

Title:Protein and fat digestibility of high and low fat diets in lean and obese mice
Abstract:
The MSc thesis aimed to investigate the effect of diet, line and sex on apparent crude protein digestibility and crude fat digestibility in FHI (lean mouse line) and FLI (fat mouse line) lines. Two types of diet, HFD (high-fat diet) and LFD (low-fat diet), were tested separately by sex and line. The digestibility experiment was performed in two replicates, at six weeks of age (1ObPr) and 13 weeks of age (2ObPr). We found that the HFD feed had a more significant effect on digestibility parameters. At the end of the experiment, males had a higher body weight than females. The HFD-FHI group of males consumed the highest crude protein and crude fat intake, whereas the HFD-FHI group of females consumed the highest crude protein intake, and the HFD-FLI group consumed the highest crude fat intake. A better apparent digestibility of crude protein was calculated in HFD-FLI and HFD-FHI females compared to the other groups. Similar findings were also obtained for crude fat digestibility, as females of HFD-FHI and HFD-FLI had better crude fat digestibility than females on LFD and males of both lines, regardless of the type of feed. Our results confirmed previous studies in this mouse model that differences in feed intake cannot explain the phenotypic differences in body fat gain and body fat percentage between the fat and lean lines. For the most part, there were no differences in this parameter, and in males, the lean line even consumed statistically significantly more feed than the fat line. However, we have demonstrated some differences between the two lines and the effect of feed composition on apparent digestibility, which should be considered in further evaluation of phenotypic differences between the two genotypes.
