Vsebnost fenolnih spojin in antioksidativni potencial kaljenih semen industrijske konoplje (Cannabis sativa L.)

Abstract
V magistrskem delu smo raziskali, kako kaljenje semen dveh sort industrijske konoplje (Cannabis sativa L.) vpliva na vsebnost fenolnih spojin in antioksidativni potencial (AOP). Semena sorte KC Dóra in Tiborszallasi smo kalili 96 h. Vsakih 24 h smo opravili vzorčenje. Odvzeta semena smo najprej razmastili, nato pa pripravili izvlečke, pri čemer smo kot ekstrakcijsko topilo uporabili 70 % etanol. Vsebnost skupnih fenolnih spojin, skupnih flavonoidov, skupnih flavonov in flavonolov in skupnih flavanonov in dihidroflavonolov, ki smo jih določili z uporabo spektrofotometričnih metod, se je med procesom kaljenja povečala. Preverili smo, kakšen AOP kažejo izvlečki semen v homogenem in heterogenem okolju. Z metodo določanja sposobnosti lovljenja radikala DPPH• in metodo določanja sposobnosti lovljenja superoksidnega anionskega radikala smo določili višji AOP v nekaljenih semenih v primerjavi s kaljenimi. Nekaljena semena so bila tudi bolj učinkovita pri preprečevanju oksidacije linolne kisline v emulziji. Trend spreminjanja AOP ni sovpadal s spreminjanjem vsebnosti skupnih fenolnih spojin. To smo pojasnili s korelacijami, ki kažejo, da so bili pri vseh treh metodah določitve AOP najuspešnejši skupni flavonoidi, ki pa so predstavljali manjši delež skupnih fenolnih spojin v izvlečkih nekaljenih in kaljenih semen.

Title:Phenolic compound content and antioxidant potential of germinated industrial hemp seeds (Cannabis sativa L.)
Abstract:
This master's thesis investigated the effect of seed germination of two varieties of industrial hemp (Cannabis sativa L.) on phenolic compound content and antioxidant potential (AOP). Seeds of KC Dóra and Tiborszallasi were germinated for 96 h. Samples were taken every 24 h. The collected seeds were first defatted, then extracts were prepared using 70% ethanol as the extraction solvent. The content of total phenolic compounds, total flavonoids, total flavones and flavonols, and total flavanones and dihydroflavonols determined by spectrophotometric methods increased during the germination process. We measured the AOP of the seed extracts in a homogeneous and heterogeneous environment. Using the method to determine the ability to scavenge the DPPH• radical and the method to determine the ability to scavenge the superoxide anion radical, we found a higher AOP in non-germinated seeds compared with germinated seeds. Non-germinated seeds were also more effective in preventing linoleic acid oxidation in the emulsion. The trend of change in AOP was not consistent with the change in the content of total phenolic compounds. This was explained by correlations, which show that total flavonoids were the most successful in all three methods of determining AOP, but accounted for a lower proportion of total phenolic compounds in the extracts of ungerminated and germinated seeds.
