Abstract
Alergije na določeno hrano so težava, s katero se sooča mnogo posameznikov, ki se morajo hrani, ki bi pri njih potencialno izzvala alergijsko reakcijo, izogibati in jo izločiti iz jedilnika. S pomočjo biotehnologije in predvsem tehnik genskega inženiringa, kot sta RNAi in CRISPR/Cas9, so se odprle možnosti pridobivanja hipoalergogenih rastlin, ki tudi pri občutljivih posameznikih ne bi sprožile nastanka alergijske reakcije. RNAi sistem je primeren za utišanje posameznih genov, ki kodirajo zapis za alergogene proteine, zato je že bil uporabljen za utišanje izražanje alergogenov v soji, paradižniku, jabolku in arašidu. V zadnjem obdobju pa vse večjo vlogo pri genskem inženiringu prevzema CRISPR/Cas9 sistem, ki je primeren za tarčno urejanje genoma in je bil uporabljen za hkratno utišanje izražanja dveh alergogenih proteinov v soji. Ključnega pomena je, da bi lahko povsem odstranili alergogene iz rastlinske prehrane, za kar je potrebno razumevanje funkcije, ki jo posamezen alergogen protein v celici opravlja. Poleg tega je potrebno določiti, v kolikšni meri so alergogeni odstranjeni iz hipoalergogene rastline, kar je mogoče s pomočjo kvantifikacije RNA in proteinov v celici, pomembna pa so tudi klinična testiranja, s katerimi se ugotavlja odziv občutljivih posameznikov ob stiku s hipoalergogeno rastlino.

Title:Plant diet allergens and biotechnological procedures for hypoallergenic plant production
Abstract:
Allergie to sertain plant based food is common problem for many people, who have to avoid foods that could potentially cause an allergic reaction and exclude them from their diet. Biotechnology and genetic engineering techniques such as RNAi and CRISPR/Cas9 offer possibility for hypoallergenic plant production, which would not trigger an allergic reaction even in sensitive individuals. Therefore they would potentially be able to consume them. The RNAi system is suitable for target silencing of gene expression that encode allergene proteins therefore it has already been used for silencing of allergene expression in soy, tomato, apple and peanut. However, recently CRISPR/Cas9 plays leading role in genetic engineering, as it is suitable for target genome editing and has been used for simultanious silencing of two soybean allergene expression. It is important to completely eliminate allergenes from plant based food, which requires an understanding of allergenic protein funcion in plant cell. Furthermore measurment of allergene reduction in hypoallergenic plant is necessary. This is possible through RNA and protein level quantification, vitally important are also clinical tests to determine response of sensitive individuals upon contact with hypoallergenic plant.
