Abstract
Mezenhimske matične/stromalnce celice izvirajo iz zarodne plasti mezoderma, iz katere se med embrionalnim razvojem razvijejo hrustanec, kostno tkivo, mišice, maščobno tkivo in druga vezivna tkiva. Najdemo jih v anatomsko posebnih področjih imenovanih niše. Niša predstavlja celično mikrookolje, ki nudi podporo in nujno potrebne dražljaje za ohranjanje samo-obnovitvenega potenciala. Zaradi imunosupresivnega delovanja se mezenhimske matične/stromalnce celice uporabljajo za zdravljenje hudih oblik bolezni presadka zoper gostitelja, globokih hrustančnih poškodb, osteoartroze in nekaterih avtoimunskih bolezni. Mezenhimske matične/stromalnce celice so lahko dostopne, saj jih lahko izoliramo iz katerega koli vezivnega tkiva odraslega človeka, imajo sposobnost adherence na plastično površino in tvorbo kolonij ter so multipotentne. Izolirane mezenhimske matične/stromalnce celice predstavljajo zelo heterogeno populacijo, zato je za pravilno identifikacijo mezenhimske matične/stromalnce celice treba upoštevati identifikacijske kriterije.Tkiva donorjev post mortem so pomemben predmet raziskav, saj predstavljajo potencialni vir različnih mezenhimskih matičnih/stromalnih celic, ki se lahko izolirajo do 17 dni po smrti. Posledično bi se z uporabo mezenhimskih matičnih/stromalnih celic izoliranih iz teh tkiv zmanjšala potreba po fetalnih ali embrionalnih virih matičnih celic, s čimer bi se izognili tudi etičnim vprašanjem. Namen magistrske naloge je določitev imunofenotipa mezenhimskih matičnih/stromalnih celic izoliranih iz različnih kolenskih (sinovijska ovojnica) in ob-kolenskih tkiv (kostno tkivo in pokostnica) po kriterijih kot jih določa Mednarodna zveza za celično terapijo. Iz vseh teh treh tkiv so bile predhodno izolirane primarne celice, ki so predstavljale naše preiskovane vzorce. Vzorcem mezenhimskih matičnih/stromalnih celic kultiviranim v pogojih in vitro smo imunofenotip določili s pomočjo pretočne citometrije, pri čemer smo uporabili protitelesa usmerjana proti površinskim označevalcem (CD45, CD14, CD19, CD73, CD90 in CD105). Ugotovili smo, da kolenska (sinovijska ovojnica) in ob-kolenska tkiva (kostno tkivo in pokostnica) donorjev post mortem predstavljajo potencialen vir celic z imunofenotipom podobnim mezenhimskim matičnim/stromalnim celicam. Dokazali smo tudi, da ni razlik v izražanju pozitivnih in negativnih označevalcev mezenhimskih matičnih/stromalnih celic med temi tremi preučevanimi tkivi donorjev post mortem. Rezultati magistrske naloge tako nakazujejo, da imata sinovijska ovojnica in pokostnica velik potencial kot tkivna vira za izolacijo primarnih celic z imunofenotipom podobnim mezenhimskim matičnim/stromalnim celicam. Kljub temu je pomembno poudariti, da velika večina naših preiskovanih celic ne dosega identifikacijskih kriterijev, ki jih predpisuje Mednarodna zveza za celično terapijo.

Title:The comparison of the immunophenotype of primary mesenchymal stem / stromal cells isolated from different tissues of the knee joint of post mortem donors
Abstract:
Mesenchymal stem/stromal cells originate from the embryonic mesoderm layer, from which cartilage, bone, muscle, fat, and other connective tissues develop during embryonic development. They are located in anatomically specific areas called niches. The niche is the cellular microenvironment that provides the support and stimulation necessary to maintain self-renewal potential. Due to their immunosuppressive action, mesenchymal stem/stromal cells are used to treat severe forms of graft-versus-host disease, deep cartilage lesions, osteoarthritis, and some autoimmune diseases. Mesenchymal stem/stromal cells are easily accessible as they can be isolated from any adult connective tissue, have the ability to adhere to plastic surfaces, form colonies and are multipotent. Isolated mesenchymal stem/stromal cells represent a very heterogeneous population, so identification criteria must be followed to properly identify the mesenchymal stem/stromal cells. Tissues from post mortem donors are an important subject of research as they represent a potential source of various mesenchymal stem/stromal cells that can be isolated up to 17 days after death. As so, the use of mesenchymal stem/stromal cells isolated from these tissues would reduce the need for foetal or embryonic stem cell sources, thus avoiding ethnical issues associated with the use of these tissues. The aim of our thesis was to determine the immunophenotype of mesenchymal stem/stromal cells isolated from different knee (synovial membrane) and the peri-knee tissues (bone and periosteum). Primary cells, previously isolated from all these tissues, presented the samples used in our study. In vitro cultured mesenchymal stem/stromal cells were immunophenotyped by flow cytometry using antibodies directed against the surface markers (CD45, CD14, CD19, CD73, CD90 and CD105). Our results are indicating that knee (synovial membrane) and peri-knee (bone and periosteum) tissues from post mortem donors represent a valuable source of primary cells with mesenchymal stem/stromal cells -like immunophenotype. Our results demonstrate that there are no differences in the expression of positive and negative mesenchymal stem/stromal cells markers between the three tissues, suggesting that the synovial membrane and the periosteum have a great potential as tissue sources for mesenchymal stem/stromal cells isolation. However, the vast majority of our isolated cells do not meet the identification criteria.
