Presnovna odzivnost astrocitov v kulturi na stimulacijo kanabinoidnih receptorjev : doktorska disertacija

Abstract
Astrociti so celice glije v centralnem živčnem sistemu s primerno lego med endotelijem in nevroni, kar jim omogoča neposreden privzem glukoze iz krvi in podporo nevronom s presnovki, kot je L-laktat. Astrociti L-laktat proizvajajo v procesu aerobne glikolize iz glukoze, ki nastane z razgradnjo glikogena. Slednji se ob adrenergični stimulaciji razgrajuje in hkrati sintetizira, ter s presnovkom L-laktata najverjetneje presnovno podpira okoliške nevrone. S poskusi, opisanimi v nalogi, smo odkrili, da ves L-laktat po adrenergični stimulaciji nastane iz glikogena. V Krebsov cikel vstopi v mirovanju dvakrat več glukoze, kot jo vstopi v aerobno glikolizo. Astrociti na svoji membrani izražajo številne z G-proteini sklopljene receptorje, ki vplivajo na presnovo, med njimi tudi kanabinoidna receptorja CB1 in CB2. Aktivacija kanabinoidnih receptorjev z agonisti povzroči stimulacijo eksocitoze, kar smo izmerili s spremljanjem membranske kapacitivnosti. Na končno povečanje L-laktata in glukoze v celici vpliva samo aktivacija CB2R, medtem ko na tranzientno povišanje koncentracije znotrajceličnega L-laktata in D-glukoze vpliva tudi aktivacija CB1R. Aktivacija obeh receptorjev nima vpliva na volumen celice ob hipoosmolarnem šoku. Zaključili smo, da kanabinoidni sistem povzroči zvečano stopnjo presnove astrocitov, zveča eksocitozo, vendar ne preprečuje nabrekanja celic po hipoosmolarnem šoku.

Title:Metabolic response of astrocytes in culture on stimulation of cannabinoid receptors : doctoral dissertation
Abstract:
Astrocytes are glial cells in central nervous system, ideally positioned to uptake glucose from the blood and to support surrounding neurons with metabolites, such as L-lactate. Astrocytes produce L-lactate in the process of aerobic glycolysis where glucose derives from glycogen degradation. Glycogen degrades after adrenergic stimulation into final product L-lactate, which may support surrounding neurons. In this work we showed that all L-lactate after adrenergic stimulation derives from glycogen, and that Krebs cycle is the main metabolic pathway in astrocytes at rest. Astrocytes express many GPCRs, including cannabinoid receptors CB1 and CB2. Activation of cannabinoid receptors with agonists stimulated exocytosis, measured with the whole-cell patch clamp membrane capacitance measurements. CB2R stimulation affected D-glucose and L-lactate metabolism, whereas CB1R only transiently increased L-lactate and glucose concentration. Stimulation of CB1 or CB2 receptors has no effect on the volume of cells at the hypoosmolar challenge. We conclude that cannabinoid signalling system affects astrocyte metabolism, stimulates exocytosis, but has no effect on the swelling of cells after hypoosmolar shock.
