Abstract
Za analizo ogljikovih hidratov je danes v uporabi mnogo različnih metod, v industrijski rabi je večina kromatografskih. V diplomskem delu opisujem kromatografsko določitev ogljikovih hidratov s fluorescenčno detekcijo. V sklopu tega sem optimiziral gradient za ločbo mešanice ogljikovih hidratov, primerjal med sabo tri različna barvila za fluorescenčno določitev ogljikovih hidratov in preveril obstojnost derivatov na sobni temperaturi in v zmrzovalniku. Za končno metodo sem izvedel tudi validacijo, kar je vključevalo preverjanje ponovljivosti, linearnosti in pa določitev meje detekcije in kvantifikacije.

Title:Determination of carbohydrates using fluorescence detection
Abstract:
There are many different analytical procedures for carbohydrate determination, most of them used in the industry are chromatographic. In this diploma thesis I discuss chromatographic determination of carbohydrates with fluorescence detection. As part of this thesis I optimized the mobile phase gradient for the separation of a carbohydrate mixture, I compared three different labels used for carbohydrate detection with fluorescence and I checked the stability of the derivatives at room temperature and in a freezer. I also performed validation of the final method, which included checking of repeatability, linearity and determination of the limit of detection and quantification.
