Abstract
V diplomskem delu, sem predstavil hidrofilno interakcijsko tekočinsko kromatografijo kot tehniko za izvedbo separacije in analize N-glikanov. N-glikani in glikani na sploh so zelo pomembna vrsta spojin v bioloških sistemih. Sodelujejo pri najrazličnejših bioloških procesih, ki so potrebni za normalno delovanje našega telesa. So tudi zelo zanimivi iz vidika zdravstva in farmacevtske industrije, saj so zelo pomembni pri razvoju novih biofarmacevtskih zdravil in služijo lahko kot biomarkerji za različna obolenja. V diplomskem delu sem opisal glikane in njihovo strukturo in razložil, zakaj so tako zahtevni za analizirat. Predstavil sem hidrofilno interakcijsko kromatografijo, njeno delovanje in mehanizem ločbe, ter mobilne in stacionarne faze, ki se pri tej tehniki uporabljajo. Obrazložil sem, kaj jo naredi tako primerno za separacijo polarnih in hidrofilnih spojin in naredil pregled različnih postopkov, kjer so uporabili HILIC za separacijo N-glikanov. Na koncu sem zastavil HILICHPLC-FL metodo za analizo 2-AB označenih N-glikanov.

Title:Separation of N-glycans using hydrophilic interactions
Abstract:
In my work I have presented the hydrophilic interaction liquid chromatography as a technique for the separation and analysis of N-glycans. N-glycans and glycans in general are an important group of compounds in biological systems. They are involved in a variety of different biological processes that are needed for the noraml function of our body. They are also very interesting for the pharmaceutical industry, as they play a crucial role in the development of new biopharmaceuticals and can be used as biomarkers for different kinds of disease. In my work I have described glycans, their structure and explained what makes them such an analytical challenge. I have presented hydrophilic interaction chromatography, how it works and its mechanism of separation and the diffrent mobile and stationary phases it uses. I explained what makes it such a good technique for the separation of polar and hydrophilic compounds and presented a couple of examples of the use of HILIC for the separation of Nglycans. At the end I have outlined a HILIC-HPLC-FL method for the analysis of 2-AB labeled N-glycans.
