Abstract
Kokulture alg in cianobakterij so pomembne zaradi študija interakcij mikroorganizmov v okolju ter potenciala za biotehnološko proizvodnjo. Eden glavnih izzivov pri delu s kokulturami je analiza sistema in določanje mehanizma interakcije med organizmi. V diplomskem delu smo predstavili teoretično ozadje kokultur alg in cianobakterij ter predlagali metodo, ki bi omogočala selektivno določanje velikosti populacij različnih alg ali cianobakterij v kokulturi. Metoda temelji na merjenju fluorescence fotosinteznih pigmentov v vzorcu kokulture in izračunu števila celic na osnovi umeritvene krivulje. Ker imajo lahko fotosintezni organizmi v celicah različne pigmente, bi s spremljanjem fluorescence takih pigmentov, po katerih se organizmi razlikujejo, lahko določili število celic posameznih vrst organizmov. Odvisnost od razlik v fotosinteznih pigmentih pa je hkrati tudi glavna slabost predlagane metode, saj potreba po uporabi organizmov z različnimi pigmenti močno omeji nabor za analizo primernih kokultur. Metoda se zanaša tudi na druge pogoje, ki morajo veljati za izbrano kokulturo, preden jo lahko analiziramo. Opisali smo preliminarne eksperimente, s katerimi lahko te pogoje preverimo.

Title:Establishment of an experimental system based on photosynthetic pigments for tracking growth of aquatic microorganisms in coculture
Abstract:
Co-cultures of algae and cyanobacteria are crucial for the study of microbial interactions in the environment and the production of valuable compounds for biotechnological purposes. One of the main challenges of working with co-cultures are the analysis of the system and the determination of the interaction mechanism. In this work, we present the theoretical background of algae-cyanobacteria co-cultures and propose a method for determining the population sizes of cyanobacteria or algae in co-cultures. The method is based on the measurement of photosynthetic pigment fluorescence and the calculation of cell number via a calibration curve. Since various photosynthetic organisms possess different pigments, specific pigment fluorescence could be used to determine the population size of a particular species. Dependence on the difference in pigments is also the proposed method's major flaw, as this requirement drastically limits the number of organisms suitable for this method. Before using the method, the selected co-culture must also meet several other conditions. We present some preliminary experiments, which can be used to determine these conditions.
