Abstract
PEGilacija je kemijska modifikacija, kjer najpogosteje pride do kovalentne vezave ene ali več poli(etilen glikol) verig na drugo molekulo. Diplomsko delo obravnava zgolj PEGilacijo in tehnike vezave poli(etilen glikol) verig na proteine, ki se uporabljajo predvsem v terapevtske namene. Na splošno se terapevtskemu proteinu poveča stabilnost, podaljša zadrževalni čas v krvi, zmanjša razgradnja s proteolitičnimi encimi in izločanje iz ledvic, zaradi česar je potrebno manj pogosto apliciranje. Poleg tega se mu izboljša topnost in zmanjša imunogenost. Večji del diplomskega dela je posvečen opisu različnih tehnik PEGilacije. Njihovo poznavanje je za uspešno načrtovanje PEGiliranih molekul ključnega pomena, saj z vsako posamezno tehniko dobimo drugače definiran produkt. Nazadnje pa delo nudi še vpogled v čiščenje PEGiliranih proteinov in njihovo karakterizacijo.

Title:Techniques of Protein PEGylation
Abstract:
PEGylation is a chemical modification where covalent attachment of one or more poly(ethylene glycol) chains to another molecule most commonly occurs. The diploma deals only with PEGylation and binding techniques of poly(ethylene glycol) chains to proteins primarily used for therapeutic purposes. In general, it increases stability, prolongs blood retention time, decreases proteolytic enzyme decomposition of the therapeutic protein and elimination of protein from the kidney, resulting in less frequent application. In addition, it improves solubility and reduces immunogenicity. The major part of the diploma is devoted to the description of various techniques of PEGylation. Their knowledge is crucial for the successful design of PEGylated molecules, as each technique gives us a differently defined product. Finally, the work provides insight into the purification and characterization of PEGylated proteins.
