Abstract
V svojem magistrskem delu sem s tekočinsko kromatografijo brez predhodne derivatizacije poskušala ločiti mešanico treh biogenih aminov; tiramina, triptamina in 2- feniletilamina. Te sem uporabila kot modelne spojine za obravnavanje biološko aktivnih spojin, ki se uporabljajo v farmacevtski industriji. Primerjala sem dva različna načina separacije. Izbrane biogene amine sem ločevala s kromatografijo ionskih parov ter kromatografijo z mešanimi režimi. Preizkusila sem štiri različne kolone in na vsaki opazovala, kako na separacijo vplivajo ionska jakost, pH in polarnost mobilne faze. Izbrane biogene amine sem zaznala s spektrofotometričnim detektorjem, saj spojine znatno absorbirajo svetlobo UV dela spektra. Na kolonah Primesep 200 in Kromosil 300-5-C18 sem razvila kromatografski metodi za separacijo in določitev izbranih biogenih aminov ter metodi validirala.

Title:Application of mixed-mode chromatography for determination of ionized pharmaceutical active compounds
Abstract:
I have studied the separation of the mixture of three biogenic amines (tyramine, tryptamine and 2-phenylethylamine) using high-performance liquid chromatography without derivatization. These amines were used as a model compounds for biologically active compounds that are often used in the pharmaceutical industry. The main goal of this study was a comparison of two different separation methods: ion-pairing and mixed-mode chromatography. On each column, I observed the effects of buffer ionic strength, pH and polarity on separation. Selected biogenic amines were determined by spectrometric detector since they all absorb the light in the UV spectrum. Corresponding chromatographic methods for separation and determination of biogenic amines were developed and validated on the columns Primesep 200 and Kromosil 300-5-C18.
